xCELLigence RTCA CardioReal time unlabeled cell analyzerMonitor myocardial cell activity for cardiac toxicity assessment. The contraction relaxation cycle of myocardial cells produces rhythmic impedance fluctuations, which are easily captured in milliseconds. Continuously monitor the intensity and periodic changes of impedance within seconds or days to evaluate the contractility and vitality of myocardial cells under various drug effects.
xCELLigence RTCA CardioReal time unlabeled cell analyzerAnalyze cells in the form of a 96 well electrode plate (E-Plate Cardio 96), place the instrument in a standard CO2 cell culture incubator, and connect it to an analysis workstation outside the incubator through a cable. User friendly software can achieve real-time control and monitoring of instruments.
characteristic
1. Provided a high-throughput method for detecting the effects of functional cardiotoxicity on short-term and long-term cardiac pulsation activity, as well as general toxicity
2. Related research on arrhythmogenic activity
3. Gain a deeper understanding of the mechanism of action of compounds
working principle
The xCELLigence RTCA Cardio system uses non-invasive label free biosensors to continuously evaluate the health status and function of myocardial cells in real-time. Inoculate myocardial cells into a microplate (E-Plate Cardio 96) with a micro gold electrode biosensor embedded at the bottom of 96 wells, and apply low voltage (less than 20 mV) to generate current between the biosensors. The current varies depending on the number of cells covering the biosensor, cell morphology, and cell adhesion strength (Figure 1A). Due to significant changes in cell morphology and adhesion strength during the contraction/relaxation cycle of myocardial cells, dynamic monitoring can be achieved (Figure 1B). The high data acquisition rate of the xCELLigence RTCA Cardio system (12.9 ms for the entire 96 well plate) provides high temporal resolution for observing subtle changes in continuous contraction/relaxation of myocardial cells. Flexible monitoring helps to study the short-term/long-term effects of compounds on the health and function of myocardial cells.

software
This user-friendly RTCA Cardio software provides excellent instrument control, enabling flexible experimental setup, data acquisition, and data analysis. This software also includes all instruments and experimental controls, which can simplify experimental setup and operation.

application
Heart disease modeling
The pathophysiological cell phenotype of genetically inherited heart disease can be modeled by disease specific human induced pluripotent stem cell cardiomyocytes (hiPSC CM). Modeling can provide a broader understanding of the mechanisms that cause damage to electrical excitation and contraction coupling.

Cardiac safety toxicology
Human induced pluripotent stem cell differentiated cardiomyocytes (hiPSC CM) have been used for physiological and disease flexibility evaluation of drugs. The xCELLigence RTCA CardioECR system provides multiple detection methods for detecting and understanding the potential toxicity of compounds and drugs.

Heart disease drug discovery
The human induced pluripotent stem cell cardiomyocytes (hiPSC CM) provide an exciting new model system for testing the efficacy and toxicity of novel therapeutic methods, including muscle strength compounds used to regulate myocardial contractility.

Comprehensive in vitro arrhythmia analysis (CiPA) plan
The CiPA program, driven by HESI, FDA, and the Society for Safety Pharmacology, aims to evaluate potential modifications to the current FDA cardiac safety assessment regulatory guidelines. XCELLigence RTCA CardioECR has been selected as the core validation technique and has participated in Phase I and II studies.
