background In cell apoptosis, chromosome DNA double or single strand breaks produce a large amount of sticky 3 '- OH ends. Under the action of deoxyribonucleotide end transferase (TdT), derivatives formed by deoxyribonucleotide and fluorescein, peroxidase, alkaline phosphatase or biotin can be labeled onto the 3' - end of DNA to detect apoptotic cells. This method is called terminal deoxyribonucleotide transferase mediated nick end labeling (TUNEL). Due to the fact that normal or proliferating cells have almost no DNA breaks, there is no formation of 3 '- OH and they are rarely stained. As a result, TUNEL has become a commonly used method for detecting DNA fragmentation (apoptosis).
