| background | Apoptosis is a fundamental biological phenomenon that refers to the autonomous and orderly cell death controlled by genes to maintain internal environmental stability. It plays a necessary role in the removal of unwanted or abnormal cells in multicellular organisms. In normal living cells, phosphatidylserine (PS) is located on the inner side of the cell membrane, while in the early stages of apoptosis, PS can flip from the inner side of the cell membrane to the surface of the cell membrane and be exposed to the extracellular environment. Annexin V is a calcium dependent phospholipid binding protein with a molecular weight of 35-36KDa, which can specifically bind to PS with high affinity. Early cell apoptosis can be easily and quickly detected by labeling Annexin V with fluorescein or biotin. Propidine iodide (PI) is a nucleic acid dye that cannot penetrate the intact cell membrane, while the cell membrane of early apoptotic cells is intact and resistant to PI. But in the late stage of apoptosis and dead cells, PI can penetrate the cell membrane and stain the nucleus. Therefore, by matching Annexin V with PI, it is possible to distinguish between cells in the early and late stages of apoptosis and dead cells. PI can be excited by 488532 or 546nm laser lines and emit red fluorescence. |
