It is also the main tool used to engulf soluble mediators of intracellular digestion, production, and release, such as cytokines, chemokines, and nitric oxide (NO). The study of the functions of various macrophage subpopulations is often hindered by the lack of reliable and specific methods for in vivo clearance of macrophages. Mechanism of action:

Clophosome ®- A (Chlorfenapyr), a negatively charged chlorophosphonic acid liposomeIt is a generation of neutral Clophosome ®- The upgraded product of N belongs to the second generation of highly efficient in vivo macrophage depletion agents, which essentially consist of large single-cell liposomes (LUVs) encapsulating chlorophosphonic acid molecules. Combined with the characteristic that free chlorophosphonic acid is difficult to penetrate the phospholipid bilayer and cell membrane of liposomes, but liposomes can be easily engulfed by macrophages. After being engulfed by Clophosome, the phospholipid bilayer of liposomes is disrupted by lysosomal phospholipase action, allowing the water-soluble drug chlorophosphonic acid inside to be released into the cell. When chlorophosphonic acid accumulates in cells and reaches the threshold concentration at which it begins to exert its therapeutic effect, the cells are irreversibly damaged and induced to die through apoptosis. The free flowing chlorophosphonic acid released by macrophages after death has an extremely short half-life in the circulatory system and is rapidly cleared in the urinary system.[See Figure 1]

If liposomes can be fully injected into the body, macrophages from different sources of the body can be cleared, including liver (Kupffer cells, Kupffer cells), spleen (different subpopulations of macrophages), lungs (alveoli but not small intestine macrophages), peritoneal cavity, lymph nodes, joints (engulfing synovial lining cells), and testes.
