Staphylococcus aureus chromogenic medium (second-generation)

The Staphylococcus aureus chromogenic medium is an improved medium developed by Qingdao Haibo Biotechnology Co., Ltd. It is used for rapid detection of Staphylococcus aureus in food, water, milk, ice cream, and meat products, with results obtained within 24 hours. Staphylococcus aureus appears as purple red colonies, while other bacteria appear as blue-green, colorless, or yellow. The vast majority of Gram negative bacteria are inhibited.

Usage:

Weigh 17.52g of this product and add 200ml of distilled water,Heat and boil 3-5 times until the agar is completely dissolved. When cooled to around 50 ℃, pour into a sterile petri dish and set aside for later use.

Operation steps:

Coating method:

1. Prepare the sample solution according to national standards, SN standards, ISO standards, or other methods;

2. Take 1ml of sample solution with appropriate dilution and add it to the center of a surface dried Staphylococcus aureus color medium plate. Make 2 plates for each dilution;

3. Use an L-shaped glass rod to apply the sample solution to the surface of the agar plate, avoiding applying it to the edge of the plate. Place the plate upright until the sample solution is absorbed by the culture medium;

4. Invert the petri dish and incubate at 36 ± 1 ℃ for 18-24 hours. The typical Staphylococcus aureus is a raised purple red colony;

5. Count all raised purple red colonies on the counting plate.

Marking method:

1. Prepare the sample solution according to national standards, SN standards, ISO standards, or other methods;

2. Take 10ul to 20ul of sample solution and streak it onto a solidified Staphylococcus aureus color medium plate;

3. Invert and incubate in an incubator at 36 ± 1 ℃ for 18-24 hours. The typical Staphylococcus aureus is a raised purple red colony;

Attention: Staphylococcus aureus microscopy showed Gram positive cocci, and the rabbit plasma coagulation enzyme test showed positive results.
