Storage of Premium Series Colony Count Test Tablets
When unopened, store at a temperature of 2-8 ℃ and use up within the shelf life. In environments with high humidity, please restore the test piece to room temperature before opening. After opening, if the test piece is not used up, it needs to be put back into the bag. The aluminum foil bag should be folded twice and sealed with adhesive tape or a sealing clip. It should be stored in the dark (not refrigerated) and used up within one month. When transporting or storing for a short period of time, place the test piece at room temperature.
Pre treatment of Premium Series Colony Count Test Samples
According to GB 4789.2-2022, 25g of solid and semi-solid samples are weighed and placed in a sterile homogenization cup or bag containing 225 mL of sterile phosphate buffer solution (or sterile physiological saline). After homogenization, a 1:10 sample homogenate is prepared; Weigh 25mL of liquid sample (or 25g when the result requires the total number of colonies per g of sample) and place it in a sterile homogenization cup or bag containing 225mL of sterile phosphate buffer solution (or sterile physiological saline). After homogenization, prepare a 1:10 sample homogenate.
Take 1mL of 1:10 sample homogenate and inject it into a test tube containing 9mL of diluent. After mixing, it becomes a 1:100 sample homogenate. Repeat this process to prepare a 10 fold series of diluted sample homogenates. For each dilution, use a 1mL sterile pipette or tip.
Select 2-3 suitable dilutions for inoculation testing based on the hygiene condition of the sample (liquid samples can also include stock solutions).
Regular environmental surfaces or sample surfaces can be sampled using a sampling rod, placed in a sampling tube, shaken and mixed evenly as a sample homogenate, and then selected at an appropriate dilution according to the hygiene condition of the sample for inoculation testing; Wetted test pieces can also be used for direct sampling.
