Sample processing: Same as GB method. Cultivation temperature: 36 ± 1 ℃. Inoculation volume: 1mL/tablet. Cultivation time: 24 ± 2h. Qualitative method: Pink single colony is positive. Sample pretreatment refers to GB4789.4-2024. Aseptically weigh 25g (mL) of the sample and add it to a sterile homogenization cup or bag preheated to 44 ° C and filled with 225mL of BPW (buffered peptone water). After homogenization, incubate at 36 ± 1 ° C for 8-18h. Gently shake the cultured sample mixture, transfer 1mL to 10mL TTB, and incubate at 42 ± 1 ° C for 18-24h. At the same time, take another 0.1mL and transfer it to 10mL RVS, and incubate at 42 ± 1 ° C for 18-24h.

After cultivation, Salmonella colonies appear pink, while non Salmonella colonies appear blue, yellow, white, or do not grow. Counting can be done visually or with a standard colony counter or other illuminated magnifying glass. For suspected bacterial colonies, the cultivation time can be appropriately extended to enhance interpretation.

Note 1: When conducting qualitative testing, dense streaking should be carried out, as single colonies are more likely to appear. 2. When removing the membrane, do not touch the detection area of the culture medium. 3. The test piece should be cultured upright, and the number of stacked pieces should not be too many. 4. Do not place the test piece directly under ultraviolet or sunlight, and do not place the test piece under fluorescent lamps for a long time. 5. Do not use contaminated test pieces. 6. After use, the test pieces should be sterilized with high-pressure steam before being processed in accordance with relevant laws and regulations. 7. A small number of needle like black spots may appear on the test piece, which is a normal phenomenon and does not affect the interpretation of the target strain. 8. After adding the sample, bubbles may appear near the anti overflow ring, which may prevent the liquid from fully diffusing. This is a normal phenomenon and does not affect the test results.
