Overview

purpose

Heparin affinity column can specifically adsorb lactoferrin. The lactoferrin in the sample is extracted by buffer solution, and the sample solution purified by heparin affinity column can be directly used for HPLC analysis. The combination of heparin affinity column and HPLC can achieve rapid determination, improve signal-to-noise ratio, and enhance the accuracy of the detection method.

Overview

Lactoferrin (Lf) is a glycoprotein mainly found in mammalian milk, with a relative molecular weight of about 80KD. Lactoferrin not only participates in iron transport in the body, but also has antibacterial, antiviral, antioxidant, and immune regulatory functions. It is considered a new type of antibacterial, anticancer drug and a food and feed additive with development potential. It has been widely added to infant and toddler food.

Each box of heparin affinity column contains 3mL of heparin affinity column, lactoferrin extraction reagent A (50 × concentrated), instruction manual, and quality control report.

Reference conditions for high performance liquid chromatography of lactoferrin

(1) Column information: C4 250mm × 4.6mm × 5 μ m, 300 Å

(2) The preliminary preparation work of the system is based on the AB flow matching ratio at the beginning of elution, and injection begins after the baseline is stable;

(3) The elution procedure for lactoferrin samples, gradient elution method: 0-5 minutes A 70%, B 30%; 5.0-10min A 45%, B 55%; 10.00-12min A 40%, B 60% 12.00-16min A 70%, B 30% A solution: 0.1% TFA; B solution: Acetonitrile.

(5) Maintenance method: After using the empty needle, run the gradient elution program to confirm that there is no lactoferrin washed out of the chromatographic column, and then clean the chromatographic column with methanol water
