Overview

EV-Save The tangential flow-through filtration (TFF) method, which is used as an efficient method for extracellular vesicle (EV) concentration and buffer replacement, is a stabilizer for extracellular vesicle preservation (TFF). TFF prevents sample accumulation on the filter membrane by flowing the sample parallel to it, minimizing sample loss as much as possible. In addition, samples such as EVs that are easily adsorbed may experience a decrease in recovery rate due to physical stress during membrane adsorption and filtration operations.

Tangential flow-through filtration (TFF) is an efficient method used for extracellular vesicle (EV) concentration and buffer replacement. TFF prevents sample accumulation on the filter membrane by flowing the sample parallel to it, minimizing sample loss as much as possible. In addition, samples such as EVs that are easily adsorbed may experience a decrease in recovery rate due to physical stress during membrane adsorption and filtration operations.

EV-Save ™ The extracellular vesicle preservation stabilizer (TFF) aims to improve EV recovery by reducing physical stress and membrane adsorption during TFF treatment. This product can achieve efficient and stable EV recycling.



◆ Characteristics


● Inhibit EV damage and EV adsorption on the membrane during EV purification, concentration, and buffer replacement using TFF

Easy to operate, only need to add this product and equilibrium buffer to the sample

This product can prevent EV loss caused by adsorption on test tubes and prevent freeze-thaw damage. The filtered (TFF) EV solution can be stored directly.

(Note: This effect can only be achieved by adding this product during the buffer exchange process)

▼ Improve the recovery rate of purified EV during buffer replacement process


Using MassivEV ™ EV Purification Column PS (product number: 131-19491) was purified from the cell culture supernatant of bone marrow-derived mesenchymal stem cells (BMMSCs) and adipose tissue derived mesenchymal stem cells (ADMSCs). Then, using Repligen's TFF system, 1/100 of the HBS buffer containing this product was replaced for buffer exchange. Using PS Capture ™ Extracellular vesicle ELISA kit (streptavidin HRP) (product number: 298-80601, using anti-CD63 or anti-CD81 antibodies for antibody detection), evaluate the EV recovery rate after buffer replacement. , The particle size distribution of EV particles was detected using Nanoparticle Tracking Analysis (NTA).

By adding this product, an increase in the recovery rate of EV based on TFF displacement buffer can be observed.

▼ Improve the recovery rate of EV purified from cell culture supernatant based on TFF

Concentrate the cell culture supernatant 20 fold using TFF, and then replace it with 100 fold buffer. Add this product in a ratio of 1/100 to the cell culture supernatant and displacement buffer. Use CD63 and CD81 ELISA kits to detect the recovery rate of EVs, and use Nanoparticle Tracking Analysis (NTA) to detect the particle size distribution of EVs.

▼ Purification of EV adsorption inhibition and cryoprotection effect


(1) Adsorption inhibition effect

Separate EV Save ™ Extracellular vesicle preservation stabilizer (product number: 058-09261, hereinafter referred to as "conventional product") and EV Save ™ The extracellular vesicle preservation stabilizer (TFF) (hereinafter referred to as "this product") was added to the purified EV solution in a dosage of 1/100. Afterwards, transfer to microtubes and store for 48 hours. Use CD63 and CD81 ELISA kits to detect the content of EVs.

(2) Freezing protection effect

Using TFF, purify EV from the culture supernatant of bone marrow-derived mesenchymal stem cells (BMMSCs). Add this product to the purified EV solution at a ratio of 1/100, and then perform 3 freeze-thaw cycles (from -20 ℃ to room temperature). Use CD63 and CD81 ELISA kits to detect EV levels before and after freeze-thaw cycles.
