Laminin is a glycoprotein with a molecular weight of approximately 900 kDa, formed by the aggregation of three sugar chains - α, β, and γ - to form a cross structure. The alpha chain, beta chain, and gamma chain each have multiple molecular types, and the type of laminin is named based on their combination (for example, the laminin composed of alpha 1, beta 1, and gamma 1 is called laminin-111).
Laminin is the extracellular matrix (ECM) that is known to be involved in cell adhesion, migration, and proliferation. Especially, laminin and type IV collagen are considered the main components of the basement membrane.
In the central nervous system, laminin is almost exclusively expressed in the basement membrane surrounding blood vessels and is used as a biomarker for blood vessels. Previous studies have shown that the inner layer (endothelial side) of the cerebral vascular basement membrane contains laminin-411 and -511, while the outer layer (brain parenchymal side) contains laminin-211 and -1111)Participate in the formation and function of the blood-brain barrier (BBB).

Protocol (Example)
1. Infusion fixation: Use 0.1% sodium citrate/PBS infusion to remove blood, followed by 4% PFA infusion
2. Rear fixation: 4% PFA (recommended for 6 hours)
3. Sucrose replacement: 30% sucrose/PBS (4 ℃, overnight~about 2 days)
4. Preparation of frozen sections: After removing sucrose, prepare frozen embedding blocks and use a low-temperature sectioning mechanism to prepare 30 μ m thick sections
5. Glycine treatment: 25 mM glycine PBS (about 20 min)
6. Closure: 5% normal goat serum, 0.3% Triton X-100/PBS (4 ℃, overnight)
7. Primary antibody: anti laminin, guinea pig (1:500), 1% normal goat serum, 0.3% TritonX-100/PBS (4 ℃, overnight~2 days)
8. Cleaning: 0.3% TritonX-100/PBS (5 min x 3 times)
9. Secondary antibody: Alexa Fluor®488 AffiniPure Goat Anti Guinea Pig IgG (H+L) (1:500) (Product Code: 568-77901),
1% normal goat serum, 0.3%TritonX-100/PBS (room temperature, 2 hours)
10. Cleaning: 0.3% TritonX-100/PBS (5 min x 3 times)
11. Packaging: Store in the dark at 4 ℃
