VGLUT (Vesicular Glutamate Transporter) is a transporter protein that transports the main neurotransmitter glutamate in mammalian excitatory synaptic transmission to synaptic vesicles. Studies suggest that it utilizes proton electrochemical gradients to transport glutamate. At present, three subtypes of VGLUT1, VGLUT2, and VGLUT3 have been found in mammals, with different expression sites. VGLUT1 is mainly expressed in the cerebral cortex and hippocampus, while VGLUT2 is expressed in the thalamus, hypothalamus, and amygdala. However, there are few neurons expressing VGLUT3. According to reports, based on mRNA expression patterns, VGLUT3 is distributed in the striatum, hippocampus, cerebral cortex, raphe nucleus, and other areas1)In the field of neuroscience, anti VGLUT antibodies are also used as labeled antibodies for glutamatergic neurons.

Protocol (Example)
1. Infusion fixation: Use 0.1% sodium citrate/PBS infusion to remove blood, followed by 4% PFA infusion
2. Rear fixation: 4% PFA (4 hours)
3. Sucrose replacement: 30% sucrose/PBS (4 ℃, overnight~about 2 days)
4. Preparation of frozen sections: After removing sucrose, prepare frozen embedding blocks and use a low-temperature sectioning mechanism to prepare 30 μ m thick sections
5. Glycine treatment: 25 mM glycine PBS (about 20 min)
6. Closure: 5% normal goat serum, 0.3% Triton X-100/PBS (4 ℃, overnight)
7. Primary antibody:Anti-VGLUT1, guinea pig (1:1600), 1% normal goat serum, 0.3% TritonX-100/PBS (4 ℃, overnight~2 days)
8. Cleaning: 0.3% TritonX-100/PBS (5 min x 3 times)
9. Secondary antibody: Alexa Fluor®488 AffiniPure Goat Anti Guinea Pig IgG (H+L) (1:1600) (Product Code: 568-77901),
1% normal goat serum, 0.3%TritonX-100/PBS (room temperature, 2 hours)
10. Cleaning: 0.3% TritonX-100/PBS (5 min x 3 times)
11. Packaging: Store in the dark at 4 ℃
