(NH ₂) 7-8ARM PEG Maleimide is an eight armed star shaped PEG, where the seven arm end is a free amino group (- NH ₂) and the one arm end is a maleimide functional group.

chemical reaction

1. Thiol specific addition (1arm Maleimide)

and SH (thiol/mercapto) undergoes Michael addition to form stable thioether bonds. The reaction is mild and often carried out under neutral to weakly alkaline conditions. It is insensitive to amino/hydroxyl groups.

2. Amine coupling/acylation (7arm Amine)

Multiple freedoms NH ₂ is a nucleophilic site that can couple or acylate/add with activated carboxyl groups (such as EDC/NHS systems, NHS esters), anhydrides, and isocyanates, forming amide bonds (amide) or urea/urethane bonds (urea/urea), respectively, suitable for multi-point grafting or crosslinking.

3. Arm function division and collaboration

The 7-arm amino group provides high connectivity for chemical fixation and network construction, while the 1-arm Maleimide provides targeted capture of thiol groups or specific anchoring of biomolecules such as Cys proteins and SH-DNA/probes. The common strategy is to first fix the material/surface with amino groups, then directionally couple the target with thiol groups or add SH first, and then crosslink at multiple points.
