NIPINon invasive prenatal screening is a method of using large-scale parallel sequencing and other technologies to deeply sequence free DNA in maternal peripheral blood and obtain fetal chromosome information. Traditional prenatal screening and diagnostic methods (early pregnancy ultrasound examination, mid pregnancy serological screening, amniotic fluid cell chromosome karyotyping analysis) have certain limitations, especially the puncture method may harm the fetus and lead to miscarriage. With the deepening of research on next-generation sequencing technology, non-invasive fetal genetic testing has developed rapidly.
CTC, also known as circulating tumor cells, typically refers to cancer cells that detach from clusters of cancer cells and enter the circulatory system;
CtDNA is called circulating tumor DNA. During the process of division and proliferation, tumor cells undergo apoptosis, death, and actively secrete DNA fragments that have undergone genetic mutations into the bloodstream. We call this DNA fragment ctDNA.

Direct extraction method
According to the description in CLSI H3-A61, samples were taken by venipuncture.
Draw blood in the order listed in CLSI H3-A61.
Fill the test tube
Remove the connecting tube from the test tube and immediately gently invert it 8 to 10 times to mix the liquid inside the tube (as shown in the figure). Insufficient mixing or delayed mixing can both lead to incorrect test results.
operating procedure
Collect blood into a test tube according to H3-A61 in the CLSI document.
After the collection is completed, please transport and store within the temperature range.
Extract cell-free DNA BCT according to the instructions of the instrument manufacturer.
Note:
The STRECK non-invasive tube does not dilute blood, therefore, there is no need to perform dilution factor correction to obtain absolute values.
As with most clinical trial specimens, hemolysis, jaundice, and lipemia can all affect the test results obtained by storing blood samples in the STRECK non-invasive tube.
Extraction of cell-free plasma DNA and cellular genomic DNA:
The extraction of cell-free plasma DNA and cellular genomic DNA can be accomplished using most commercially available assay kits.
To obtain results, a proteinase K treatment step (≥ 30 mAU/ml) is added to the 60 ℃ free solution salt. It takes 1 hour to extract cell-free DNA and 2 hours to extract cellular genomic DNA.

【 Precautions 】

3. Please use within the validity period of the test tube.
4. Test tubes cannot be used for collecting injectable materials.
5. Do not dilute or add other ingredients.
6. Excessive or insufficient filling of test tubes can cause an imbalance in the ratio of blood and additives, leading to incorrect analysis results or reduced product performance.
7. Warning
Glass may break, please take preventive measures during the handling process.
All biological specimens and materials in contact with the product will be considered as infectious biological hazards, and should be handled in accordance with relevant local regulations. Avoid contact with skin and mucous membranes.
This product should be disposed of as infectious medical waste.
When removing or reinstating the test tube stopper, gently shake the stopper or grip it while twisting and pulling outward. Do not use your thumb to push out the stopper, as it may cause the test tube to rupture and cause injury.
8. Prevent backflow
Due to the presence of chemical additives in the STRECK non-invasive tube, efforts should be made to prevent liquid from flowing out of the tube.
To prevent backflow, please follow the following preventive measures:
Please keep the patient's arm in a downward position during the collection process.
Try to hold the stopper upwards when holding the test tube.
Blood begins to flow into the test tube or the tourniquet is released two minutes after use.
During the collection process, objects inside the test tube should not come into contact with the stopper or needle.
STRECK Non Invasive Tube Storage and Transportation
At temperatures between 18-30 ℃, the STRECK non-invasive tube can be stored stably within its shelf life.
Do not refrigerate empty STRECK non-invasive tubes. Appropriate isolation measures should be taken during transportation under temperature conditions.
Blood samples collected in the STRECK non-invasive tube for cell-free DNA analysis can be stored stably for 14 days in an environment of 6-37 ℃.
