SHIMSEN QuEChERS Ceramic Proton Centrifuge Tube Dispersion Extraction

Nantong Hairuo Chemical Co., Ltd

The process of using ceramic grinding or dispersing components to process the sample and then transferring it to a centrifuge tube for separation.

A brief overview of QuEChERS method

QuEChERS (short for "fast, simple, economical, efficient, durable, and safe") is a sample pretreatment method proposed by the NONG industry in the United States in 2003.

This method has been widely applied in the pretreatment of various samples, especially for the pretreatment of pesticide residues.

Compared to traditional methods, it is simpler, more economical, and faster.

The six characteristics of QuEChERS method:

Quick: The extraction and purification steps are time-consuming.

Easy: With fewer operating steps, there is no need for complex equipment.

Cheap: Low solvent usage and low cost of consumables.

Effective: High recovery rate of analytes and good purification effect.

Rugged (durable/reliable): The method has good tolerance and is not easily affected by matrix interference.

Safe: The use of organic solvents (usually acetonitrile) is minimal and relatively environmentally friendly, resulting in minimal harm to operators.

DSPE, short for Disperse Solid Phase Extraction, is the core purification step in the QuEChERS method.

The QuEChERS method uses acetonitrile or acidified acetonitrile as the extraction solvent. Excess salts or buffer salts are added to the extraction solution for liquid-liquid extraction, followed by centrifugation. The acetonitrile layer is transferred to a purification tube containing anhydrous sulfuric acid MG and purification reagent for mixing, further removing impurities and water from the extraction solution. Centrifuge and collect the supernatant to complete the pretreatment step.

AOAC 2007.01 Method

1. Accurately weigh 15g of homogeneous sample, add 15mL of 1% acetic acid acetonitrile solution (V/V), add QuEChERS extraction salt package 6g MgSO4+1.5g NaOAc, and internal standard solution.

2. Shake vigorously for 1 minute, centrifuge>1500x g, 1 minute.

3. Transfer 1mL or 8mL of supernatant to a dSPE purification tube, vigorously shake for 1 minute, centrifuge>1500x g for 1 minute.

[EN 15662 method]

1. Accurately weigh 10g of homogeneous sample ①, add 10mL of acetonitrile and internal standard solution, and vigorously shake for 1 minute. (If the moisture content of the sample is less than 80%, water must be added before homogenization, please refer to EN15662:2008 (E) 5.2 ②)

2. Add QuEChERS extraction salt package 4g MgSO4+1g NaCl+1g TSCD+0.5g DHS ③ to the above extraction solution, shake vigorously for 1 minute, centrifuge>3000x g for 5 minutes.

3. Transfer 1mL or 6mL of supernatant to a dSPE purification tube, shake vigorously for 1 minute, centrifuge>3000x g for 5 minutes.

Note:

① The sampling amount of the sample depends on the sample matrix, fruit and vegetable samples, with a sampling amount of 10g ± 0.1g; grain and honey samples, with a sampling amount of 5g ± 0.05g; tea and spice samples, with a sampling amount of 2g ± 0.03g

② When the moisture content of the sample is less than 80%, sufficient cold water (<4 ° C) must be added before homogenization of the sample. Please refer to EN15662:2008 (E) 5.2 for the moisture content of various common samples and the amount of water that needs to be added

③ TSCD Sodium Citrate, DHS Sodium Hydrogen Citrate

If necessary, the above two methods can be diluted, nitrogen blown, or solvent replaced for GC/MS-MS or LC/MS-MS analysis.

PSA mainly removes interference from sugars, fatty acids, organic acids, anthocyanins, and other substances in the sample matrix;

C18 mainly removes lipids and non-polar interfering substances from the sample matrix;

GCB (graphitized carbon) mainly removes pigments, sterols, and non-polar interference.

[Explanation 2]

A 2mL purification tube is suitable for transferring 1mL of extraction solution;

A 15ml purification tube is suitable for transferring 6-8mL of extraction solution.

The role of homogeneous protons in ceramics

1. Excessive anhydrous MgS0 needs to be added during the extraction and purification process, as excess salt is prone to clumping. Adding ceramic protons can effectively prevent salt clumping.

2. During oscillation and centrifugation, it can accelerate the diffusion of adsorbents and better extract and purify samples.
