PrimocinTMAseptic guardians of organoid research

Organoids, a guest of honor in CNS journals, rose to fame and were widely reported by various media outlets

In 2009, the Hans Clevers team published a groundbreaking achievement in the journal Nature, which was the first in vitro cultivation of colorectal epithelial organoids;

In 2013, it was named one of the top ten technologies of the year by Science magazine;

In 2015, it was listed as one of the top ten technological breakthroughs by MIT Technology Review;

In 2018, it was named the 2017 Method of the Year by Nature Methods;

In 2019, it was evaluated by The New England Journal of Medicine as an excellent human preclinical disease model.

From 2023 to present, a milestone in organoid research has been achieved, with thousands of papers related to organoid research published

Nowadays, at the forefront of life science and medical research, organoid technology is reshaping our understanding of human development, disease modeling, and personalized treatment in unprecedented ways. These three-dimensional organ models, which are cultured, miniaturized, and functionalized in vitro, provide us with a unique window to explore the mysteries of life. Moreover, due to the characteristics of short time consumption, high throughput, and ability to preserve the heterogeneity of human tissue, organoid models have high application value.

However, this promising technology has faced a tricky challenge of microbial contamination since its inception.

Organoids refer to tissue analogues with a certain spatial structure formed by in vitro three-dimensional (3D) culture of adult stem cells or pluripotent stem cells. Organoids are not truly human organs, but they can simulate real organs in terms of structure and function, simulate tissue structure and function to the greatest extent possible, and can be stably cultured for a long time.

The cultivation process of organoids is delicate and complex. It usually comes from precious stem cells or tissue biopsy samples and requires careful cultivation in nutrient rich media for weeks or even months. During this period, any accidental contamination by mycoplasma, bacteria, or fungi could be catastrophic for researchers

1. Precious sample loss:Tumor organoids or primary tissue samples from patients are usually not obtainable;

2. Huge waste of time and resources:Weeks of hard work, expensive growth factors, and culture media may be wasted due to overnight pollution;

3. Unreliability of experimental results:Invisible pollution (such as mycoplasma) may not immediately kill cells, but it can quietly alter the gene expression, proliferation, and differentiation behavior of organoids, leading to biased experimental data and invalid conclusions.

Traditional antibiotics such as penicillin -/streptomycin (dual antibiotics) are widely used, but their effectiveness is often limited and they cannot comprehensively prevent all microorganisms. Long term use may also be toxic to cells themselves, affecting the normal growth and function of organoids.

PrimocinTM: Protect your precious cells and prevent cell culture contamination

PrimocinTMIt has always been used as a tool to prevent contamination in primary cell culture, because primary cells derived from animals and tissues are highly susceptible to contamination by isolated hosts and microorganisms in the environment. Primocin ™ Antibiotics can effectively prevent contamination by bacteria, mycoplasma, and fungi, while minimizing cell toxicity. Now it has gradually become a trusted "guardian angel" in the field of organoid research.

Primocin provided by InvivogenTMContains four compounds, three of which inhibit DNA and protein synthesis in Gram positive bacteria, Gram negative bacteria, and mycoplasma. The fourth compound eliminates fungi, including yeast, by disrupting ion exchange on the cell membrane. Primocin ™ It has been successfully used in many primary cell lines, including mouse and human tumor cell lines, embryonic cells, and induced pluripotent stem cells.

Core advantages:

efficientPrevent mycoplasma, bacteria, fungi, and yeast;

Reference citationVerified by Nature Protocols stem cell research;

Safe and non-toxicSpecially designed for primary cells. At an effective bactericidal concentration, there is no significant negative impact on the self-renewal, proliferation, and differentiation and maturation of stem cells and organoids;

cost-effective1mL PrimocinTMCan prepare 500mL cell culture medium;

Easy to operatePrimocinTMIt can be directly added to the culture medium, with a simple operation process. Cultivating cells at the recommended concentration can have a preventive effect.

PrimocinTMApplication scenarios of organoid related research:From basic scientific research to the forefront of clinical practice

Construction of primary organoids:In the initial stage of constructing organoids from tissue fragments, the sample itself may carry microorganisms, PrimocinTMThe use of is crucial as it can effectively 'purify' the starting materials and improve the success rate of construction.

Long term subculture:For tumor organoid or stem cell banks that require long-term expansion and stability, continuous use of Primocin is recommendedTMA sturdy firewall can be established to ensure the purity and security of cellular resources.

Drug screening and sensitivity testing:Ensuring that all organoids are not contaminated is a prerequisite for obtaining accurate and reproducible efficacy data when used for high-throughput drug screening. PrimocinTMProviding confidence assurance for such critical applications.

Regenerative Medicine and Future Therapies:With the gradual application of organoids in cutting-edge fields such as transplantation and gene therapy, the sterile requirements for culture materials have reached drug level. The high standards of PrimocinTM have cleared the way for such applications.
