OCT embedding agent

OCT frozen section embedding agent is a water-soluble mixture of polyethylene glycol and polyvinyl alcohol, which is widely used in immunohistochemistry laboratories. Its purpose is to support tissues during frozen section to increase tissue continuity, reduce wrinkles and fragmentation. Also, as the OCT mixture is water-soluble, it is soluble in water during the bleaching process, so background staining will not be added in future staining.

Using OCT mixture (optim um cutting temperature compound) to pre infiltrate tissues, and then performing constant cold box sectioning to improve the quality of sectioning.

OCT embedding agent(SAKURA product, MADE IN USA) embedding process:
1. Remove the tissue block from the sucrose solution and add 20g L-1 sucrose to itOCT embedding agent1: Soak in a mixture of 1 volume ratio at room temperature for 2 hours
2. Transfer to OCTEmbedding agentSoak at room temperature for 4 hours and replace with a new oneOCT embedding agentSoak at room temperature for 6 hours
3. Place the specimen on a tray and slice it using a constant cold box slicer (usually -22 ℃) with a thickness of 10 μ m. Attach it to a pre treated glass slide and store it in a -20 ℃ freezer.
