Cell Description
Synovium is the inner layer of the joint capsule, light red, smooth and shiny, thin and soft, composed of loose connective tissue. All structures within the joint cavity, except for articular cartilage and meniscus cartilage, including tendons, ligaments, etc. that pass through the joint cavity, are enveloped by synovium.
Synovium secretes synovial fluid, which plays an important role in joint movement. The normal synovium is divided into two layers, namely the thin cell layer (lumen layer) and the vascular layer (subendometrium layer). It is the vascular rich inner membrane of the joint capsule, attached to the non articular surface, covering the bone surface inside the joint capsule, and not on the cartilage surface. This part is called the marginal zone or "bare zone". The synovium is pink, smooth, shiny, wet, and lubricating, sometimes with visible villi containing collagen fibers.
Synovial cells have two types, A and B. Macrophage like type A cells have filamentous pseudopodia, serosal invagination, vesicles, mitochondria, lysosomes, cytoplasmic fibers, and Golgi apparatus on their surface, and possess phagocytic function; B-type fibroblast like synovial cells (FLS) have a high concentration of endoplasmic reticulum structure and are the main cells mediating RA joint destruction.
Cell characteristics
1) The cells are derived from normal joint tissue of experimental animals.
2) Cell identification: Vimentin immunofluorescence staining was positive.
3) After identification, the purity of the cells was found to be higher than 90%.
4) Does not contain HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi.
5) Cell growth mode: fibroblast like cells, adherent culture.
Transportation and storage of products
Depending on the weather conditions and transportation distance, the company and the customer will negotiate and choose one of the following methods.
1) 1mL cryopreserved cell suspension is put into 1.8ml cryopreserved tube and transported in foam insulation box filled with dry ice; After receiving the cells, please thaw and revive them for cultivation as soon as possible. If resuscitation cannot be performed immediately, the frozen cells can be stored at -80 ℃ for one month.
2) The T-25 culture bottle is filled with culture medium and transported at room temperature; After receiving the cells, please observe the growth status of the cells under a microscope. If the bottle laying rate exceeds 85%, immediately perform passaging operation. If there are many suspended cells, please place the culture bottle in the incubator overnight to help non dead suspended cells adhere to the wall again.
