GeneCopoeia in the United States provides target expression clones based on the action of * genomic miRNAs, covering public databases. The miRNA target expression clone contains the downstream 3 '- UTR (miRNA action site) sequence of the reporter gene (firefly luciferase) driven by the SV40 promoter. The second reporter gene, Renilla luciferase, can be used to monitor transfection, expression efficiency, and can serve as an internal reference.

Characteristics of Human Genome miRNA Target Expression Cloning

By analyzing the activity of firefly luciferase, the regulatory effects of miRNA on potential target genes can be obtained. After expression cloning and transcription, a chimeric mRNA of firefly luciferase ORF and 3'UTR is formed. If there is a specific miRNA that can complement and bind to a certain segment of the 3'UTR sequence, the activity of the protein encoded by the luciferase ORF in the chimeric protein is also regulated accordingly. Therefore, by detecting the luminescence intensity of luciferase, the mRNA regulation effect of miRNA corresponding to the target can be quantitatively determined.
