This product is used for quantitative determination of human luteinizing hormone in human serum or plasma (anticoagulated with heparin lithium).

This product is an automatic quantitative test used on VIDAS series analyzers, using ELFA technology (enzyme-linked fluorescence analysis) to quantitatively detect human luteinizing hormone in human serum or plasma (heparin lithium as anticoagulant). Luteinizing hormone (LH) is a glycoprotein with a molecular weight of approximately 30000 daltons. It has two peptide subunits (alpha and beta) that are linked together by non covalent bonds to form LH. The alpha subunit of luteinizing hormone (LH) consists of 92 amino acids and is identical to the alpha subunit sequences of follicle stimulating hormone (FSH), thyroid stimulating hormone (TSH), and human chorionic gonadotropin (hCG). The beta subunit consists of 121 amino acids and contains the amino acid sequence of *, which gives luteinizing hormone biological and immune specificity. Luteinizing hormone molecules also include carbohydrates, accounting for 20% of the molecular weight of the hormone. The luteinizing hormone (LH) is secreted by the anterior pituitary gonadotropin cells in a pulsatile manner and controlled by the hypothalamus. Luteinizing hormone stimulates the production of steroid hormones (testosterone for males and estradiol for females), which have a negative feedback effect on gonadotropins. In males, luteinizing hormone stimulates Leydig cells to produce testosterone. In menstrual women, the synergistic effect of luteinizing hormone and follicle stimulating hormone directly affects the development of follicles, increases their steroid products, and determines ovulation. During menopause, the production of estradiol sharply decreases, leading to an increase in serum levels of luteinizing hormone. Whether or not combined with follicle stimulating hormone, the luteinizing hormone (LH) assay is an important parameter for studying reproductive function. Elevated levels of luteinizing hormone in males and amenorrhea women indicate primary hypogonadism, while lower levels of luteinizing hormone indicate secondary hypogonadism (hypogonadotropic hypogonadism). 5,6,7

The analysis principle adopts a combination of enzyme immunoassay sandwich analysis method and final fluorescence detection (ELFA).

Solid phase tube (SPR) ®) In analysis, it serves not only as the solid phase surface for reaction, but also as a pipetting device. The reaction reagent is ready to use and is added to the well of the reagent strip in advance, and then the well is sealed. All analysis steps are automatically performed by the instrument. The reactants enter and exit the SPR several times.

Firstly, add the sample to a well containing alkaline phosphatase labeled anti luteinizing hormone (LH) antibody (conjugate). The sample/conjugate mixture is circulated in and out of SPR several times to increase the reaction rate. The antigen binds to the antibody coated on SPR and binds to the conjugate to form a 'sandwich'. Unrelated components are eliminated during the rinsing step.

In the final detection step, the substrate (4-methylumbelliferone phosphate) is circulated in and out of the SPR. The conjugate enzyme catalyzes the hydrolysis of the substrate into a fluorescent product (4-methylumbelliferone), and its fluorescence intensity is measured at 450nm. The fluorescence intensity is directly proportional to the concentration of antigens present in the specimen. After the detection is completed, the instrument calculates the results based on the calibration curve stored in the memory, and then prints them out.
