This product is used for in vitro quantitative detection of human chorionic gonadotropin in human serum or plasma (heparin lithium or EDTA). Quantitative detection of human chorionic gonadotropin in human serum or plasma (heparin lithium or EDTA) using enzyme-linked fluorescence analysis (ELFA) technology. Human chorionic gonadotropin (hCG) is a glycoprotein with a molecular weight of 4000 daltons. HCG is composed of two peptide subunits, alpha and beta. About 30% of the weight of hCG is composed of polysaccharides (hexose, sialic acid). Structurally, hCG is similar to the three pituitary hormones LH, FSH, and TSH. The alpha subunit (92 amino acids) is almost identical among the four hormones mentioned above. The beta subunit is composed of 145 amino acids and 5 oligosaccharide groups, and contains an amino acid sequence of *, which endows hCG with biological and immunological specificity [1,2]. However, only α - β dimers have biological activity [3,4,5,6]. Human chorionic gonadotropin (hCG) is mainly synthesized by placental syncytiotrophoblast cells. The level of serum hCG steadily increases and reaches its peak in the first three months of pregnancy, and then gradually decreases until the sixteenth week of pregnancy, after which it remains stable throughout the entire pregnancy. The physiological function of hCG is to maintain the corpus luteum during the first three months of pregnancy and promote the production of progesterone and estrogen. HCG also plays a role in the differentiation of the fetal reproductive tract. HCG testing is crucial in the diagnosis and follow-up of pregnancy (ectopic pregnancy, molar pregnancy, etc.) [7,8]. HCG can also be produced by certain tumor tissues, mainly those originating from trophoblast cells (choriocarcinoma, etc.). In this case, hCG acts as a biomarker and is a crucial clinical indicator for prognosis and treatment monitoring [9].

The analysis principle adopts a combination of enzyme immunoassay sandwich analysis method and final fluorescence detection (ELFA). Solid phase tube (SPR) serves not only as the solid-phase surface for antigen antibody reactions in analysis, but also as a pipetting device. The reaction reagent is ready to use and is added to each well of the reagent strip in advance, and then each well is sealed. All analysis steps are automatically performed by the instrument. The reactants enter and exit the solid-phase tube (SPR) several times. Firstly, take a sample and add it to the well containing anti hCG antibody labeled with alkaline phosphatase. The sample/conjugate mixture enters and exits the solid phase several times during operation to increase the reaction rate. The antigen binds to the antibody encapsulated in the solid-phase tube and binds to the conjugate to form a "sandwich". Then, the conjugate in the fifth well of the reagent strip is circulated into and out of the solid-phase tube, and the unbound hCG sites are saturated. Unrelated components are eliminated during the rinsing step. In the final detection step, circulate the substrate (4-methylumbelliferone phosphate) multiple times inside and outside the solid-phase tube. Conjugated enzyme catalyzes the hydrolysis of the substrate into a fluorescent product (4-methylumbelliferone). The fluorescence of the product was measured at 450 nanometers. The fluorescence intensity is directly proportional to the concentration of antigens present in the sample. At the end of the analysis, the instrument automatically calculates the results based on the calibration curve stored in the memory by VIDAS, and then prints them out.
