VIDAS T3 is an automated quantitative assay used on VIDAS series analyzers, utilizing ELFA technology (enzyme-linked fluorescence analysis) to quantitatively detect total triiodothyronine (T3) in human serum or plasma (anticoagulated with heparin lithium). Triiodothyronine (T3) is a hormone secreted by the thyroid gland (20%), and a portion of T3 (80%) is produced by peripheral deiodination mechanisms that convert T4 to T3. The physiological activity of T3 is much stronger than T4, therefore it plays an important role in maintaining normal thyroid function.
T3 circulates in the form of free hormones (0.3%) or binds to carrier proteins such as TBG, albumin, or prealbumin (>99.7%). The free form has physiological activity and has the greatest effect on metabolic control. T3 must be analyzed in conjunction with other tests, such as TSH and FT4, as well as clinical examinations of patients.
The analysis principle adopts a combination of enzyme immunoassay competitive analysis method and final fluorescence detection (ELFA).
Solid phase tube (SPR) ®) In analysis, it serves not only as the solid phase surface for reaction, but also as a pipetting device. The reaction reagent is ready to use and is added to the reagent strip in advance, and then the reagent strip is sealed. All analysis steps are automatically performed by the instrument. The reaction reagent enters and exits SPR several times. Firstly, take a sample and add it to a well containing T3 antigen labeled with alkaline phosphatase (conjugate). The antigen present in the sample will compete with the labeled antigen to bind to the anti-T3 antibody (derived from sheep) encapsulated on the inner surface of SPR. Unrelated components are eliminated during the rinsing step. In the final detection step, the substrate (4-methylumbelliferone phosphate) circulates in and out of the SPR. Conjugated enzyme catalyzes the hydrolysis of substrate into fluorescent product (4-methylumbelliferone), and its fluorescence intensity is measured at 450nm. The fluorescence intensity is inversely proportional to the concentration of antigens present in the specimen. At the end of the analysis, based on the calibration curve stored in the memory, VIDAS automatically calculates the results and then prints them out.
