This product is a product available in VIDAS ® The automatic quantitative reagent used on the series analyzer uses ELFA (enzyme-linked fluorescence analysis) technology to quantitatively detect total tetraiodothyronine (T4) in human serum or plasma (heparin lithium as anticoagulant) using enzyme immunoassay. Thyroxine or tetraiodothyronine (T4) is a hormone secreted by the thyroid gland, with the majority (99.9%) binding to its carrier protein. These carrier proteins are mainly TBG (thyroid hormone binding globulin). A small amount of free thyroxine is considered the active form of this hormone. This product helps evaluate the function of the thyroid gland. In patients with hyperthyroidism, thyroid hormone levels are elevated, while in patients with hypothyroidism, thyroid hormone levels generally decrease.
The detection of T4 depends on its concentration in the carrier protein, so it is necessary to test the binding ability of hormones to the carrier protein. The detection of this binding ability also needs to be combined with other thyroid function assessments such as TSH and T3, as well as clinical examinations of the patient.
The thyroid hormone detection kit (enzyme-linked fluorescence assay) is helpful for the diagnosis of thyroid dysfunction.
【 Principle of Inspection 】
The thyroid hormone detection kit (enzyme-linked fluorescence method) adopts the analysis principle of combining enzyme immunoassay competitive analysis method with final fluorescence detection (ELFA).
Solid phase tube (SPR) ®) In analysis, it serves not only as the solid phase surface for reaction, but also as a pipetting device. The reaction reagent is ready to use and is added to the well of the reagent strip in advance, and then the well is sealed.
All analysis steps are automatically performed by the instrument. The reactants enter and exit the SPR several times.
Firstly, take a sample and add it to a well containing T4 antigen (conjugate) labeled with alkaline phosphatase. The antigen present in the sample competes with the labeled antigen to bind to the specific anti-T4 antibody coated on the inner surface of SPR.
In the final detection step, the substrate (4-methylumbelliferone phosphate) circulates in and out of the SPR. The conjugate enzyme catalyzes the hydrolysis of the substrate into a fluorescent product (4-methylumbelliferone), and its fluorescence intensity is measured at 450nm. The fluorescence intensity is inversely proportional to the concentration of antigens in the sample.
At the end of the analysis, based on the calibration curve stored in the memory, VIDAS ® The analyzer automatically calculates the results and then prints them out.
