The specific manufacturing process of earthworm fibrinolytic enzyme freeze-dried powder is as follows:

1) Making Earthworm Coarse Powder: Starve live earthworms to naturally spit out sediment, clean them, air dry them, crush them at low temperature, and then sieve them to make earthworm coarse powder;

2) Preparation of crude enzyme solution: Mix the earthworm powder evenly with phosphate buffer PBS and sodium chloride solution, and place it in a 2-8 ℃ freezer for 12 hours. 3) Purification and modification of crude enzyme: the crude enzyme solution of earthworm fibrinolytic enzyme was subjected to Sephadex G75 gel chromatography, and then was eluted by washing solution to obtain the pure enzyme of earthworm fibrinolytic enzyme. Then add activated PEG modifier and boric acid buffer for insulation reaction, purify the supernatant through phosphate buffer dialysis and centrifugation, and collect the active ingredients to obtain earthworm fibrinolytic enzyme modifier;

4) Freeze drying of earthworm fibrinolytic enzyme: Load earthworm fibrinolytic enzyme modified enzyme into a tray with a material thickness of about 8mm, freeze at -45 ℃ for 300min, and then dry once under a vacuum of 35pa with a temperature control range of -30~10 ℃, sublimation drying time of 720min, remove most of the flowing water, and then perform decomposition drying with a temperature control range of 10~40 ℃ and a drying running time of 860min. According to the endpoint judgment of the Chinese biological freeze-drying machine, the freeze-drying process is completed to obtain earthworm fibrinolytic enzyme freeze-dried powder with a water content of less than 1%.
