Used for serum typing of suspected Escherichia coli strains identified by biochemical tests,
This product is a serum containing antibodies against Escherichia coli. It undergoes agglutination test with the corresponding serotype of Escherichia coli and is used for serum typing of Escherichia coli.
Main components (see Appendix for details)
1. Escherichia coli serum (1mL/bottle x 51 bottles): a total of 40 types, used for serotype typing of pathogenic, invasive, toxic, and hemorrhagic Escherichia coli. 2. Pathogenic Escherichia coli serum (1mL/bottle x 18 bottles): a total of 15 types of serum, used for serotype typing of pathogenic Escherichia coli; 3. Invasive Escherichia coli serum (1mL/bottle x 11 bottles): consisting of 11 types of serum, used for serotype typing of invasive Escherichia coli: 4. Toxic Escherichia coli serum (1mL bottle x 10 bottles): consisting of 10 types of serum, used for serotype typing of toxic Escherichia coli: 5. Hemorrhagic Escherichia coli serum: Hemorrhagic Escherichia coli serum O157, 1 bottle: Escherichia coli serum H7, 1 bottle; Hemorrhagic Escherichia coli serum O104, 1 bottle: Escherichia coli serum H4, 1 bottle.
Clean glass slides or reaction plates (cards), inoculation rings (rods) that are required but not provided.
【 Storage conditions and expiration date 】
Stored in the dark at 2-8 ° C, with a validity period of 18 months from the date of self inspection and qualification.
【 Sample Requirements 】 1. The sample needs to undergo preliminary biochemical testing and be suspected of being a strain of Escherichia coli. 2. The strains that have undergone biochemical testing should be cultured on ordinary nutrient agar medium at 35-37 ℃ for 18-20 hours to obtain fresh bacterial cells that can be used for serum agglutination tests.
Inspection method
The tested bacteria were first subjected to agglutination tests with OK multivalent serum, 0157 and 0104 serum, respectively; If agglutination occurs with OK multivalent serum, perform agglutination test with the corresponding monovalent serum in the multivalent serum (see Appendix for multivalent corresponding monovalent serum): If agglutination occurs with a certain monovalent serum, prepare a thick bacterial solution of the tested bacteria, heat it in a 100 ℃ water bath for 30 minutes, and then perform slide agglutination test with the corresponding monovalent serum. If it is still positive or negative, the bacterial type can be determined. If the test result is O157 or 0104, then the bacteria (not heated) will undergo agglutination reaction with H serum. If there is no agglutination with H serum or the agglutination strength is weak, the bacteria can be transferred to soft agar medium and cultured at 35-37 ℃ for 18-20 hours, and then subjected to agglutination test with H serum. (Using pathogenic, invasive, and toxic Escherichia coli serum kit, ignore the steps related to 0157 and O104 serum in this step. Using hemorrhagic Escherichia coli serum, ignore the steps related to OK multivalent serum in this step.) Operation method: Drop 1 drop of serum on a clean glass slide, then mix a small amount of the tested bacteria with the serum, gently shake the glass slide, and visually judge the result within 1 minute
