OCT embedding agent (SAKURA product) embedding process:

1. Remove the tissue block from the sucrose solution and immerse it in a 1:1 volume ratio mixture of 20g L-1 sucrose and OCT embedding agent at room temperature for 2 hours

2. Transfer to OCT embedding agent and soak at room temperature for 4 hours. Replace with a new OCT embedding agent and soak at room temperature for 6 hours

3. Place the specimen on a tray and slice it using a constant cold box slicer (usually -22 ℃) with a thickness of 10 μ m. Attach it to a pre treated glass slide and store it in a -20 ℃ freezer.

OCT frozen section embedding agent is a water-soluble mixture of polyethylene glycol and polyvinyl alcohol, which is widely used in immunohistochemistry laboratories. Its purpose is to support tissues during frozen section to increase tissue continuity, reduce wrinkles and fragmentation.
