When using an anaerobic incubator, it is necessary to strictly follow the installation, operation, and maintenance specifications to ensure a stable anaerobic environment and ensure experimental safety.
Anaerobic incubator is a key equipment used for cultivating anaerobic microorganisms, and its core is to maintain a constant, anaerobic environment. To prevent oxygen infiltration from causing cultivation failure or bacterial death, preventive measures need to be taken at multiple stages.
Preparation and installation precautions before operation
Install the instrument in a place with clean air and minimal temperature changes as much as possible to avoid external environmental interference.
Ensure that the power socket is reliably grounded, and the equipment is strictly prohibited from being transported or handled upside down.
Before starting up, it is necessary to fully familiarize oneself with the instructions for using the supporting instruments and meters, and master the correct operating methods.
Check if the pneumatic system is intact and regularly inspect for any air leaks.
Key steps and points in the operation process
The main process and precautions for creating an anaerobic environment:
1. Initial replacement (nitrogen): Two replacements of the operating room with nitrogen require controlling the exhaust through a foot switch to ensure sufficient gas replacement
2. Mixed gas displacement input: N ₂ (85%), H ₂ (10%), CO ₂ (5%). The mixed gas flow rate is controlled at about 10 milliliters per minute and slowly released into the operating room
3. Catalytic deoxygenation: After starting the palladium particle deoxygenation agent and catalytic converter power supply for one hour, use Meilan indicator to detect the color change. If there is no color change, the anaerobic state is reached
4. Sample transfer: Enter and exit the sample through the sampling chamber, first evacuate and then fill with nitrogen. Close the inner and outer doors tightly, and if necessary, use low vacuum to test the sealing
The culture must be placed in an anaerobic environment in the operating room before it can be placed, otherwise it may cause the bacteria to be exposed to oxygen and die
