Introduction: 4T1 is a 6-thioguanosine resistant cell line obtained from the 410.4 tumor strain without mutagenesis screening. When injected into BALB/c mice, 4T1 spontaneously generates highly metastatic tumors that can metastasize to the lungs, liver, lymph nodes, and brain, while forming an initiating lesion at the injection site. It is not necessary to remove the initial lesion when inducing metastasis.

The growth and metastasis characteristics of 4T1 mouse breast cancer cells in BALB/c mice are very similar to those of human breast cancer cells. This tumor is an animal model of human stage VI breast cancer. The dynamics of tumor metastasis induced by 4T1 are similar between postoperative and non-surgical conditions, and can be used as a model for both postoperative and non-surgical conditions. Compared with other tumor models, small metastatic cell clusters (as few as one) can also be detected in many distant organs due to the anti-6-thioguanosine property of 4T1.
