Do not neglect the use and maintenance of chromatographic columns,Assist in the development of laboratory methods with half the effort

Switching from positive phase mode to negative phase mode:

Rinse the chromatography column with 20 times the column volume of methanol/ethanol (isopropanol) (90:10, v/v). Use a new mobile phase activated chromatography column with at least 20 times the column volume and a low flow rate of 0.3ml/min

If the salt of the buffer in the reverse phase is insoluble in methanol and/or ethanol, rinse the chromatography column with a suitable solvent (such as 50/50 methanol/water or 50/50 acetonitrile/water), and then activate the chromatography column with a new mobile phase

Once the chromatographic column is switched to reverse phase condition, it is not recommended to switch back to normal phase condition

preservation:

NP: n-hexane/isopropanol (90:10, v: v)

PO: Acetonitrile/isopropanol (95:5, v: v) or methanol/isopropanol (95:5, v: v)

RP: Methanol/water or acetonitrile/water, and the water ratio should always be below 20%

Regeneration:

Rinse the chromatography column with 100% ethanol or methanol (40 ℃) at an appropriate flow rate for 1-3 hours
