【 Principle of Inspection 】

This reagent kit uses colloidal gold immunochromatographic double antibody sandwich method to detect the presence of antibodies in specimensVibrio cholerae01, 0139 group antigens（Ag）， Using colloidal gold as a marker and nitrocellulose membrane as a coating carrier. The nitrocellulose membrane is coated with two types of antibodies, one of which is antiVibrio cholerae01 or 0139 groupMonoclonal antibodies (Ab) are used to capture antigens in specimens; Another type is sheep anti mouse multi antibody, used for quality control of this test strip. Combined with other raw materials, assemble into test strips. During the detection process, the specimen is moved by the chromatographic action of the microporous membrane, and antigen antibody reactions occur during the movement. If the specimen containsVibrio cholerae01 or 0139 groupFirstly, it binds with colloidal gold labeled antibody (Au Ab) to form an Au Ab Ag complex. When the Au Ab Ag complex encounters the antibody coated on the membrane, it forms an Au Ab Ag Ab double antibody sandwich complex, presenting a visible red band.

Sample processing:

• Direct testing: Water samples are directly tested.2. Bacterial enrichment detection

Water sample stool:Inoculate 1-2 ml in 8-9 ml alkaline peptone water and incubate at 37 ℃ for 6-8 hours or overnight.

Solid fecal specimens: Inoculate 1-2g in 8-9ml alkaline peptone water, mix well, and incubate at 37 ℃ for 6-8 hours or overnight.

1. Before testing, the sample and test strip should be equilibrated to room temperature;

2. Place the test strip on a clean and dry workbench;

3. Take 2-3 drops of the sample using the pipette provided in this kit or a clean sample dispenser, and add them to the sample addition end of the test strip (white end of the test strip) to start timing;

4. Observation results for 2-15 minutes, reading results for 15 minutes or more is invalid.

Explanation of Inspection Results

Negative: A red line appears, indicating the presence of a red line at the position of the quality control line;

Positive: Two red lines appear, indicating the presence of red lines at both the detection and quality control lines;

Limitations of Inspection Methods

Because this reagent is effective for 104Bacteria with a concentration of less than/ml cannot be detected. When determining the result, only qualitative judgment can be made for positive or negative, and no quantitative judgment can be made.
