The following are the specific sterile techniques for cell line culture:

Before the experiment begins, the sterile room and sterile operating table need to be irradiated with ultraviolet light for 30 to 60 minutes for sterilization. The sterile operating table should be wiped with 70% ethanol, and the sterile operating table fan should be turned on and run for 10 minutes before the experiment can proceed. Each operation can only handle one cell line, and even if the culture medium is the same, it cannot be shared to avoid contamination or confusion between cells. After the experiment is completed, please take the experimental items out of the workbench and wipe the sterile operating table again with 70% ethanol. The operation interval should allow the sterile operating table to run for 10 to 15 minutes before proceeding to the next cell line operation.

2. The sterile operation area should be kept clean and spacious, and necessary items (test tube racks, straws, pipettes, or straw boxes, etc.) can be temporarily placed. Other experimental supplies should be removed after use to facilitate air circulation. Experimental supplies need to be wiped with 70% ethanol before being brought into the sterile operating table. The experimental operation process should be completed in the central sterile area of the table, and should not be performed in non sterile areas at the edges.

3. Be careful when using sterile experimental materials to avoid bacterial contamination. Do not touch the tip of the straw or the mouth of the container, and do not operate directly above the opened container. After opening the container, please hold the bottle cap with your hand and gently grip the bottle body. Tilt it at an angle of about 45 degrees to use, and try not to place the bottle cap upside down on the table.

4. Staff should first pay attention to their own safety and must wear laboratory clothes and gloves before conducting experiments. Special care should be taken when handling viral infections or cell lines from humans, and appropriate levels of sterile workstations should be selected (at least Class II). During the operation, it is necessary to avoid the production of aerosols, be careful of toxic drugs such as DMSO and TPA, and avoid needle injuries, etc.

5. Regularly check the following items:

5.2. CO2 concentration, temperature, and contamination of the water tray in the CO2 incubator (sterile water is required for the water tray, and it should be replaced regularly every week).

5.3. The airflow pressure inside the sterile operating table requires regular replacement of UV lamps and HEPA filter membranes, as well as pre filters (300 hours/pre filter, 3000 hours/HEPA).

6. Disinfectant (Zephrin 1:750) can be added to the sink, and the water in the sink needs to be replaced regularly.
