Experimental principle and steps

1. Design and selection of siRNA: Select appropriate databases and software to screen siRNA targeting sequences based on research objectives. Typically, multiple siRNAs need to be designed to identify the target sequence*excellent*Effective siRNA sequences;

2. Preparation of siRNA: Common in vitro preparation methods include chemical synthesis, in vitro transcription, degradation of long dsRNA by RNase Il, and expression of siRNA in cells using siRNA expression plasmid vectors, viral vectors, and siRNA expression frameworks (SECs) prepared by PCR;

3. siRNA transfection: siRNA. The main methods for transfection into cells using siRNA expression vectors or siRNA expression frameworks include calcium phosphate co precipitation transfection, electroporation transfection, and mechanical methods (microinjection and gene gun) Transfection with cationic liposome reagent;

4. Detection of target gene expression: WB and other methods can be used to detect the expression of target genes, or their expression can be analyzed by labeling siRNA (such as fluorescent labeling). It can also be used for siRNA intracellular localization and double labeling experiments to track the transfection process and target gene expression silencing process.
