Ni agarose gel H.P. is a purification medium used to purify 6 × His tag recombinant protein. It is made by coupling a tetradentate chelator NTA with 6% cross-linked Sepharose It can be used to purify any expression system expressed 6 × His tagged recombinant protein under non denaturing or denaturing conditions. NTA， Containing four chelating regions, it can better bind Ni2+than typical three tooth chelating agents. 6 × His can chelate with Ni2+, allowing His tagged protein to bind to Ni NTA purification medium. Unbound protein is washed off, and protein bound to the medium is gently eluted with a certain concentration of imidazole or low pH buffer to obtain high-purity target protein. The purification medium has an affinity of * with His tag protein, reaching 5-20 mg/mL. The His tag protein obtained from any expression system can be purified under both non denaturing and denaturing conditions. The purification process is simple, and the purity of the obtained protein can reach up to 95%. Ni NTA can be regenerated 4-6 times and reused. The suspension of this product is 20% ethanol, which has been chelated with Ni2+.
