Micro aerobic anaerobic incubator YQX-IIMain features:
1. The anaerobic chamber consists of a cultivation operation room, a vacuum sampling room, an air circuit, a circuit control system, and other parts. The cultivation room adopts a hanging cabinet style, which increases the usage space of the operating room. Therefore, it is possible to configure some small instruments according to the requirements of internal control in the operating room, such as the Corning oscillator. We have customized them for multiple universities according to this requirement.
2. The anaerobic incubator adopts scientific and simple methods to achieve high-precision and constant temperature anaerobic environment, making it easy for operators to obtain an anaerobic environment and operate and cultivate anaerobic bacteria in the anaerobic environment.
3. The temperature control adopts a digital temperature controller, which can accurately and intuitively set the temperature you need and reflect the actual temperature inside the box. With effective temperature limiting protection settings, it is safe and reliable. The incubator is equipped with ultraviolet disinfection lamps, which can sterilize and disinfect at any time. After gas filtration, it enters the box, which can effectively prevent bacterial contamination.
4. The pneumatic device can adjust the flow rate. The aisle room and operating room are both made of stainless steel plates. The front window of the anaerobic incubator is made of transparent and impact resistant special glass plate, and is equipped with an operating hole locking device to more effectively ensure the gas concentration inside the incubator.
1、 Preparation before operation
1. Check the gas source
Confirm that the pressure of nitrogen, hydrogen, and carbon dioxide cylinders is sufficient (usually ≥ 0.3MPa), and the gas purity is ≥ 99.99% to avoid impurities affecting the anaerobic environment.
Check if the gas pipeline connection is sealed and free of leaks (can be detected with soap water).
2. Pre reduced culture medium
Before using the culture medium, it needs to be boiled to remove oxygen and quickly placed in an anaerobic chamber for pre reduction for more than 24 hours (if using a cutting sky blue indicator, ensure a colorless state).
3. Pre operation of equipment
Turn on the power of the anaerobic tank in advance and run it for at least 2 hours (or as required by the manual) to ensure that the tank reaches an anaerobic state (oxygen concentration<1ppm, which can be verified with an oxygen indicator).
2、 Precautions for operation process
1. Sample transfer specifications
Use dedicated transfer cabin (transition cabin):
First evacuate (to 0.1MPa), then fill with nitrogen (repeat 2-3 times) to ensure that oxygen is replaced.
Avoid frequent opening of the transfer cabin and restore anaerobic environment after each operation.
2. Personal protection
Wear powder free gloves (to avoid powder contamination of the indoor environment), and use masks and goggles if necessary.
Open flames or static electricity are strictly prohibited when operating gases such as hydrogen.
3. Key points for operating inside the box
Tools and items need to be disinfected with alcohol and dried before being placed to avoid introducing oxygen or moisture.
Gently move, reduce airflow disturbance, and avoid glove damage (regularly check glove sealing).
Containers such as culture dishes and test tubes need to be sealed (such as using anaerobic bags or stoppers).

3、 Post use maintenance
1. Cleaning and disinfection
Regularly wipe the inner walls, gloves, and trays of the box with 75% alcohol or specialized disinfectant to prevent microbial contamination.
Waste liquid and culture medium should be removed in a timely manner to avoid the growth of miscellaneous bacteria.
2. Gas management
After closing the gas cylinder valve, it is necessary to release the residual pressure in the pipeline (to prevent backflow).
When not in use for a long time, turn on the machine at least once a week to maintain an anaerobic environment.
3. Catalyst regeneration
Palladium catalyst (deoxidizer) is regenerated by baking at 160 ℃ for 2 hours every 23 months, and replaced promptly after failure.
4、 Safety Warning
1. Hydrogen risk
Hydrogen is flammable and explosive. Ensure that the box is well grounded and operate away from sparks and high-temperature equipment.
2. Risk of hypoxia
Avoid inserting your head into the box to prevent suffocation (the box is in an anaerobic environment).
3. Emergency response
In case of gas leakage or fire, immediately turn off the gas source and use a carbon dioxide fire extinguisher (do not use water or dry powder).
5、 Common problem handling
High humidity inside the box: Add silicone desiccant or activate dehumidification function.
Elevated oxygen concentration: Check the sealing of gloves, whether the operation steps of the transfer chamber are standardized, or replace the catalyst.
Cultivate contamination: Check whether the disinfection procedures of the transfer chamber and the sterilization of the culture medium are effective.
