MTS Cell Proliferation Detection KitThe MTS Cell Proliferation Assay Kit is a common cell proliferation and toxicity detection kit. MTS is a novel tetrazolium salt that can be reduced by succinate dehydrogenase in live cell mitochondria to the water-soluble product formazan. Methylenediamine has specific absorption at 490 nm, so cell proliferation can be reflected by measuring the absorbance at 490 nm. The faster the cell proliferation, the higher the absorbance value.
Compared with common MTT assay kits, this kit does not require the removal of the original culture medium or the use of additional solvents to dissolve formazan, making it more convenient to use and avoiding experimental errors caused by removing some formazan from the culture medium. This reagent kit is very flexible to use and can be further incubated after detection to obtain better color results. In addition, this reagent kit also has the advantages of wide linear range, high sensitivity, and safe use.

Usage
Experimental operation:
1. MTS working fluid preparation:
Mix MTS Solution and PMS Solution in a 20:1 ratio to obtain MTS working fluid. If the prepared MTS working fluid needs to be stored, it should be kept at -20 ℃ away from light. At this time, it is recommended to divide it appropriately to avoid repeated freezing and thawing.
Attention: MTS Solution and PMS Solution are prone to decomposition under light, so they need to be protected from light during preparation and use (can be wrapped in tin foil). If frequent use is required and this kit needs to be stored at 4 ° C, MTS working solution should be prepared before each test. The prepared and unused MTS working solution needs to be stored at -20 ° C in the dark.
2. Cell culture:
Inoculate the cells into suitable containers such as well plates. For 96 well plates, it is recommended to add 100uL of culture medium to each well, and it is also recommended to inoculate 5-100 × 10 cells per well3A cell. According to the experimental design, interested drugs can be used to treat the experimental group cells for a period of time. At this time, a blank group (containing only the culture medium) and a control group (containing the culture medium, cells, and drug solvent) need to be established separately.
Note: The specific number of cells to be inoculated in each well can be adjusted according to the specific cell type.
3. MTS incubation:
Add 20 uL of MTS working solution to each well, and then incubate in a cell culture incubator for 1-4 hours.
Attention: The incubation time of MTS can be adjusted according to specific experiments.
4. Testing:
Measure the absorbance (A) at 490 nm using an enzyme-linked immunosorbent assay (ELISA) reader. If there is no 490 nm filter, you can choose to use a 450-540 nm filter.
5. Data analysis:
The formula for calculating cell survival rate is as follows
Cell survival rate (%)=[(A)experimental group-Ablank group) / (Acontrol group-Ablank group)] ×100
Precautions
1. When cells are cultured for a long time, the edge holes of the 96 well plate are prone to liquid evaporation problems. Suggest discarding the area around the 96 well plate and adding sterile water, culture medium, or PBS instead; At the same time, the 96 well plate can be placed in the incubator near the water source.
2. MTS Solution and PMS Solution will solidify at low temperatures. Before use, please warm them back to room temperature or a 37 ℃ water bath for about 10 minutes until they are completely melted before use.
3. MTS Solution and PMS Solution are sensitive to light. If stored at 4 ° C, the color of the solution may change slightly, which may increase the background absorbance value but will not affect the final experimental results.
4. If it cannot be detected in a timely manner, 25uL of 10% SDS can be added to each well after incubation in step 3 to terminate the reaction. The orifice plate processed by SDS can be placed
