The homogeneous ready to use 3D cell viability detection kit is used to quantitatively detect the ATP content in 3D model cells, and the ATP content is directly proportional to the number of live cells. This reagent has the characteristics of high signal-to-noise ratio, good repeatability, and good stability. The ready to use formula reduces the experimental steps of cracking before detection, further reducing errors caused by frequent sample addition. | Usage |
1. Cell preparation:
(1) 3D cell model construction: Construct relevant 3D cell models according to experimental requirements, such as 3D cell sphere models. (2) If the experiment is to determine the effect of a compound on a 3D cell model, add an appropriate concentration of the compound to be tested to the cell plate wells. The concentration of organic solvents in the culture medium should be kept below 2%. Continue to cultivate suitable time according to experimental requirements. 2. Cell viability testing: (1) Remove CTG-LTM3D Cell viability assay kit , equilibrate to room temperature (22 ℃ -25 ℃) [Note 2], shake gently and mix well. (2) Remove the test cell culture plate and equilibrate to room temperature (22 ℃ -25 ℃). 3D cell viability testing needs to be performed on a white experimental plate. If the cell culture plate to be tested is transparent or black, the 3D model needs to be transferred to the white experimental plate for testing. (3) Add 50 µ L of 3D cell viability assay reagent to 100 µ L of 96 well plate 3D model cells, or 10 µ L of reagent to 20 µ L of 384 well plate 3D model cells . (4) Vigorously shake the experimental plate for 5 minutes to fully lyse the cells [Note 4], and place it in the dark to equilibrate the reaction for 25 minutes. (5) Read the fluorescence signal on the luminescence reader [Note 5]. |

| Precautions |
1. After the second use of the reagent, it should be packaged and stored in the dark at -20 ℃ or below to ensure the stability of the reagent. The signal intensity of the 3D cell viability detection reagent decreased by less than 10% after repeated freezing and thawing for 5 times without any loss of function. It was left at room temperature (22 ℃) for 8 hours or 4 ℃ for 72 hours with a signal intensity decrease of less than 10% and no loss of function.
2. The luciferase reaction in 3D cell viability detection reagents is sensitive to temperature changes. The reagents and experimental plates need to be equilibrated to room temperature (22 ℃ -25 ℃), and the temperature during the testing process should be kept constant (± 1 ℃). 3. It is not recommended to change the dosage of reaction reagents without strict verification. The volume ratio of cell culture medium to detection reagent should be 2:1. 4. It is necessary to vigorously oscillate the experimental plate to fully lyse cells, while also ensuring that the liquid in the experimental well does not overflow. 5. The 3D cell viability detection reagent has different attenuation rates for 3D model signals of different cell types, with signal half lives ranging from 1.5 hours to 3 hours. It is recommended that the plate reading time not exceed 1 hour to 2 hours. |
