Introduction to Experimental TechniquesLong non coding RNA (IncRNA) is a type of RNA with a transcript length exceeding 200 nt and does not encode proteins. IncRNA was initially considered as the "noise" of genome transcription and did not have biological functions. However, recent studies have shown that IncRNA can regulate gene expression at the epigenetic, transcriptional, and post transcriptional levels, and is involved in various important regulatory processes such as X chromosome silencing, genomic imprinting, chromatin modification, transcriptional activation, transcriptional interference, and nuclear transport. It is closely related to the occurrence, development, and prevention of human diseases.
Although more and more IncRNAs have been discovered, the current understanding of IncRNAs is only the tip of the iceberg, and the functions of the vast majority of IncRNAs are still unclear. At present, functional research on IncRNA is mainly conducted through methods such as IncRNA chips or next-generation sequencing. At present, the development of gene chip technology is becoming mature and stable. On this platform, different IncRNA probes can be designed to quickly and high-throughput screen for differentially expressed IncRNA and mRNA information in diseases or specific biological processes, and ultimately identify the target gene loci of IncRNA for in-depth analysis of regulatory mechanisms.

BioPulverizerTM frozen crushed tissue was added with 1m RNA extraction reagent TRIzol (Invitrogen) and homogenized with Mini-Bad-Bater-16 to extract RNA.
2. The experimental subjects are cell samples, with 1x106~1x107 cells taken from each sample, and 1 meter of RNA extraction reagent TRIzol added (the sample is adherent cells, and the amount of TRIzol used per 10cm2 culture dish is 1 meter). RNA is extracted after lysis.
