Product Description
Lipopolysaccharides, also known as LPS, are a component of the cell wall of Gram negative bacteria (G -). They are located on the outermost layer of the cell wall and are exposed to the cell surface of non enveloped bacteria. They help maintain the integrity of the cell outer membrane and protect bacteria from damage caused by bile salts and lipid antibiotics. LPS is composed of lipid A, core polysaccharides, and O-polysaccharide side chains. Lipids A are the main components of bacterial endotoxins, determining their strength. The high degree of variation of O-polysaccharide side chains among different xi bacteria determines the specificity of xi bacterial serotypes.
LPS can cause a cascade reaction of immune stimulation and toxic pathophysiological activities in the body, including the release of endotoxins leading to septic shock and peripheral vascular collapse. Meningitis is often diagnosed clinically by detecting the presence of LPS. Usage
1. Preparation of LPS storage solution (related to cell culture): Resuspend 1mg of LPS in 1ml of sterile equilibrium solution or cell culture medium, gently vortex and shake until the powder is completely dissolved, resulting in 1mg/ml storage solution. This storage solution can be further diluted with sterile equilibration medium or cell culture medium to the desired working concentration.
2. Preservation of LPS storage solution: This storage solution (1mg/ml) can be stored at 2-8 ° C and remains stable for about a month; It can also be divided into single doses and stored at -20 ℃ for up to 2 years of stability. Avoid repeated freezing and thawing.
[Attention]:
1. LPS storage solution should be stored in a silanization container, as LPS can adsorb onto plastic or certain glassware, especially when the concentration is less than 0.1mg/ml. But when the concentration of LPS is greater than 1mg/ml, the above-mentioned adsorption loss can be ignored.
If using glassware, vortex oscillation of LPS storage solution should be carried out for at least 30 minutes to fully dissolve the adsorbed solute.
Precautions
1) Due to the fact that LPS is a micelle forming molecule in any solvent, LPS solutions dissolved in water and phosphate are slightly turbid to turbid. Organic solvents will not make the LPS solution clearer. The dissolution of Jia Chun produces a chaotic suspension with suspended particles. However, water dissolution produces a homogeneous turbid liquid.
