J2 monoclonal antibody (mAb J2) is a mouse derived IgG2a antibody that targets double RNA (dsRNA). Its recognition ability plays an important role in virus detection and research. The J2 antibody was initially developed for the detection of plant viruses, but with further research, its application scope rapidly expanded. In 2006, Weber et al. confirmed in a study that J2 antibodies can detect a large amount of dsRNA produced by all positive stranded RNA viruses in infected cells. This discovery has made J2 antibody the "gold standard" for detecting dsRNA and has driven its widespread application in related fields, with over 200 related literature published so far.

J2 antibody can recognize dsRNAs with a length greater than or equal to 40 bp, and its recognition process does not depend on the sequence or nucleotide composition of the antigen. After research, it was found that J2 antibody can recognize various naturally occurring dsRNAs, including hepatitis C virus, dengue virus, rhinovirus, chikungunya virus, rabies virus, poliovirus, swine fever virus, and sparrow straw mosaic virus. This makes J2 antibody a tool in virological research. This antibody is suitable for various experimental techniques, including enzyme-linked immunosorbent assay (ELISA), dsRNA immunoblotting, immunoaffinity chromatography, and immunohistochemistry in certain systems. It should be noted that before dsRNA immunoblotting, nucleic acid separation must be performed by polyacrylamide gel electrophoresis, because the sensitivity after transfer from agarose gel will be significantly reduced. In addition, J2 antibodies are not suitable for clinical use and are limited to in vitro experiments.

These studies have laid the foundation for the widespread application of J2 antibodies.
