Operation steps:
Extraction of Solid Tissue Proteins
Add 10 μ L phosphatase inhibitor, 1 μ L protease inhibitor, and 5 μ L 100mM PMSF to every 1mL of cold LysisBuffer and mix well. Store on ice for a few minutes and set aside.
Every 100mg of solid tissue is placed in a culture dish, cut into small pieces of about 3mm × 3mm, add 0.5-1mL of cold LysisBuffer, and manually homogenize 15 times with a glass homogenizer, paying attention to low-temperature operation;
Transfer the tissue homogenate to a 1.5mL pre cooled centrifuge tube, centrifuge at 10000 rpm, and centrifuge at 4 ℃ for 5 minutes;
Transfer the supernatant to a new pre cooled centrifuge tube to obtain the whole protein extract, and quantify the protein using Bradford and BCA methods;
Pack and store at -70 ℃ to avoid repeated freezing and thawing.
Cultivate cell protein extraction
After suctioning off the culture medium, the cells cultured on the wall were washed twice with 10mL/150mm cold PBS and shaken several times each time to remove the culture medium as much as possible;
Suspension cultured cells or adherent cells scraped off with a cell scraper, transfer the cells and culture medium to a centrifuge tube, centrifuge at 1000 rpm for 10 minutes, and wash twice with cold PBS in a 10mL/150mm culture plate at 1000 rpm for 5 minutes;
Add 10 μ L phosphatase inhibitor, 1 μ L protease inhibitor, and 5 μ L 100mM PMSF to every 1mL of cold LysisBuffer and mix well. Store on ice for a few minutes and set aside.
After cell washing, transfer to a new pre cooled centrifuge tube and add the prepared cold LysisBuffer.
Place on a 4 ℃ shaker platform and gently oscillate for 15 minutes;
14,000rpm， Centrifuge at 4 ℃ for 15 minutes, obtain the supernatant as the whole protein extract, and quantify the protein using Bradford and BCA methods;
Pack and store at -70 ℃ to avoid repeated freezing and thawing.
