Experimental operation process for customized protein chip detection service
Introduction to Experimental Techniques:
Protein chipIt is a high-throughput monitoring system that monitors the interactions between protein molecules through the interaction between target molecules and capture molecules. Capture molecules--Usually pre fixed on the chip surface, it is widely used as a capture molecule due to the high specificity and strong binding properties of antibodies to antigens. It is crucial to effectively immobilize antibodies on the surface of protein chips, especially in terms of antibody immobilization and antigenicity, in order to enhance the sensitivity of protein chips. G protein is an antibody binding protein that specifically binds to antibody FC fragments, and has therefore been widely used to immobilize different types of antibodies.
Experimental technology principle:
The basic principle of protein chip technology is to immobilize various proteins in an orderly manner on various carriers such as filter membranes and glass slides to become detection chips. Then, antibodies or other components labeled with specific fluorescein are used to react with the chip. After cleaning, the components that cannot complement the protein on the chip are washed away. Then, fluorescence scanners or laser confocal scanning techniques are used to measure the fluorescence intensity of each point on the chip. The interaction between the components on the chip and the components in the test solution is analyzed by fluorescence intensity to achieve the measurement purpose.
Experimental operation process:
1. Customers provide biological samples (antigens or antibodies) for acceptance.
2. Prepare the sampling solution and use the Smartarrayer sampling instrument for sampling.
3. The chip is fixed, and if there are no subsequent experiments, the chip will be delivered to the customer.
4. Chip reaction experiment.
5. Chip scanning and data analysis. After chip cleaning, scan with LuxScan 10K-A Cy3 channel and extract data for analysis.
6. Provide a result report (including hybridization images, data results, and corresponding clustering and differential analysis results).
The customer provides:
1. The experimental subjects are tissue samples. Take an appropriate amount (50-100mg) of fresh tissue samples or correctly stored tissue samples, and use them
BioPulverizerTM frozen crushed tissue was added with 1m RNA extraction reagent TRIzol (Invitrogen) and homogenized with Mini-Bad-Bater-16 to extract RNA.
2. The experimental subjects are cell samples, with 1x106~1x107 cells taken from each sample. Add 1m of RNA extraction reagent TRIzol (the sample is adherent cells, and the amount of TRIz0I used per 10cm2 culture dish is 1ml), lyse and extract RNA.
