Dualex is a new multifunctional blade measuring instrument developed by PESSL, an Austrian company (formerly Force-A, a French company), based on technology from the French National Academy of Sciences (CNRS) and the University of Paris XI. It can accurately measure the chlorophyll content of leaves, as well as the flavonoid and anthocyanin content in the surface layer of leaves, and is suitable for research in plant physiology and agronomy (such as chlorophyll concentration in rice, nitrogen status in corn, grapevine, etc.).

The measurement object can be monocotyledonous plants, dicotyledonous plants, or perennial plants. This device is simple and easy to use, capable of real-time and non-destructive measurements. Due to the absence of calibration and pre sample preparation, measurement work can be completed in the laboratory or on-site. In addition, the device can operate under various temperature and ambient lighting conditionsPerform simple, fast, and non-destructive measurements of chlorophyll, polyphenols, and anthocyanins in leaves.

Accurate measurement of chlorophyll content

Chlorophyll plays a crucial role in photosynthesis and plant development. This device measures chlorophyll by analyzing the light projected onto the leaves. The system has undergone chemical calibration and the measured value is µ g/cm ² (within the range of 5-80 µ g/cm ²). Clip design sensor - measuring polyphenols and anthocyanin polyphenols in leaves mainly synthesized after receiving light. Therefore, it is actually a good indicator of the history of plant light interaction. This device measures polyphenols and anthocyanins by analyzing their shielding effects on chlorophyll fluorescence. The content of polyphenols and anthocyanins is displayed by relative absorption units: polyphenols, 0-3; Anthocyanins, 0-1.5

NBI ®: Nitrogen balance index

Chlorophyll is often used as an indicator of plant nitrogen status. Years of research and experiments have shown that polyphenols, especially flavonols, are also good indicators of plant nitrogen status.

NBI ® The nitrogen balance index combines chlorophyll and flavonols with nitrogen/carbon partitioning. This index is an indicator of plant nitrogen status and is directly related to the abundance of nitrogen elements. Compared with chlorophyll fluorescence (leaf age, leaf thickness), NBI ® The nitrogen balance index is insensitive to changes in environmental conditions.

Principle of polyphenol measurement

Chlorophyll infrared fluorescence (2) is measured by reference excitation light (1) that is not absorbed by polyphenols; Compared with the results of measuring polyphenols using light (such as green light (3) reflecting anthocyanins, or ultraviolet light (4) reflecting flavonoids), due to the absorption of polyphenols, only a small portion of the light reaches chlorophyll in the leaf flesh and can produce infrared light.

Chlorophyll measurement principle

The chlorophyll content in leaves can be quickly measured by the transmittance of light. The first beam of near-infrared light (5) is used to measure the chlorophyll content in the leaves, and the second beam of near-infrared light (6) measures the interference value of leaf structure on chlorophyll content.

The chlorophyll absorption rate of leaves is measured based on the transmittance of two types of near-infrared light. Two beams of near-infrared light (710nm and 850nm) directly irradiate the leaves, and the chlorophyll absorption rate of the leaves is calculated by comparing the transmittance detected by the detectors.

The Nitrogen Balance Index (NBI) is the ratio of chlorophyll (CHL) to flavonoids (FLAV): when nitrogen fertilizer stress does not occur, plants grow healthily, synthesize more chlorophyll, and produce fewer polyphenols (flavonoids); When nitrogen fertilizer stress occurs, plant nutrition is imbalanced, producing more polyphenols (flavonoids) and less chlorophyll. In traditional methods, only chlorophyll is used to determine nitrogen fertilizer status. When the chlorophyll content in leaves decreases (leaves turn yellow), it indicates that the plant is lacking nitrogen fertilizer. In fact, this method has a certain delay effect, and the decrease in chlorophyll is a manifestation of nitrogen fertilizer deficiency several days or even ten days ago. Even if fertilizer is applied at this time, it will affect the final yield of crops. Through this new index, earlier and more targeted nitrogen information about crops can be obtained.

When using the nitrogen balance index to evaluate nitrogen fertilizer status, the delay effect in traditional methods is avoided. A slight change in chlorophyll and polyphenols (flavonoids) can detect the nitrogen fertilizer status of plants and enable timely and rapid nitrogen fertilizer management.
