According to the PAT guidelines, dissolved carbon dioxide (DCO ₂) is a key process parameter (CPP) in the biopharmaceutical production process. It affects different metabolic pathways involved in cell growth, product formation, and quality by influencing other parameters such as extracellular and intracellular pH.
Dissolved carbon dioxide (DCO) in biopharmaceutical applications2)Traditionally, monitoring has been done through indirect pH measurement (Westinghouse technique) - requiring operators to address multiple drifts through product calibration and electrode maintenance multiple times. Most importantly, embedded pH sensors must be replaced frequently. To simplify this process, HAMILTON BONADUZ's CO2NTROL solid-state sensors use a novel optical method to directly measure dissolved carbon dioxide values. Carbon dioxide molecules diffuse into the gas permeable membrane, and the sensor measures the absorption of specific mid infrared wavelengths of carbon dioxide, which is directly related to the partial pressure of carbon dioxide (liquid or gas) in the sample.
The result is that the measurement is more accurate than traditional sensors, requires no maintenance, does not need to replace parts, and can automatically control the dissolved carbon dioxide content in real time. These improvements also ensure reproducibility across different batches, reduce costs, and improve consistency throughout the entire processing process - from research and development to large-scale production.
