【 Detection Principle 】

The DENV Detect IgM ELISA kit is a two-step enzyme-linked immunosorbent assay system. In this experiment, dengue fever negative control, positive control, and unknown concentration serum samples were diluted with sample diluent, and then added to micropores coated with anti human IgM antibodies for incubation. Afterwards, dengue fever heat source recombinant antigen DENVRA and normal cell antigen (NCA) were added separately for incubation. After incubation and plate washing, treat with HRP labeled DEN specific antibodies. After the second incubation and washing of the plate, TMB substrate solution was added for incubation. Afterwards, add acidic termination solution and measure the OD value at 450nm to determine the degree of substrate reaction. When it exceeds a certain value (critical value), the absorbance value of the sample can determine the presence of dengue antibodies in the sample.

Sample preparation: Collect serum, plasma, or whole blood samples from suspected infected individuals and preprocess them according to the instructions.

Reagent preparation: Take out the required reagents, dilute and prepare according to the instructions.

Sample addition: Add the processed sample and reagent into the microplate and follow the instructions in the manual.

Incubation: Place the microplate with added samples and reagents in a constant temperature incubator for a certain period of time.

Cleaning: Use cleaning solution to clean the microplate and remove unbound substances.

Color development: Add substrate color development solution and observe the color changes in the microplate.

Result interpretation: Determine the presence of dengue virus IgM antibodies in the sample based on the colorimetric results.
