Packaging: R1 is 3 bottles x 0.5 mg freeze-dried powder, R2 is 3 bottles x 12 mL liquid
Principle: When platelet rich plasma (PRP) is added, collagen binds to platelets through GPIa/IIa and GPVI receptors. GPVI receptors are responsible for platelet activation, leading to the release of ADP and thromboxane A2, as well as the activation of calcium ions in platelets. The response of platelets to collagen typically exhibits a latency determined by collagen concentration, followed by morphological changes manifested as a decrease in transmittance, and finally a simple aggregation waveform. Collagen is considered a potent agonist.
📌 purpose
1. Used for in vitro quantitative detection of platelet aggregation in human citrate plasma using light transmission aggregation (LTA) method.
2. Assist in diagnosing platelet dysfunction or evaluating the efficacy of antiplatelet drugs (such as P2Y12 inhibitors).
3. Suitable for laboratory professionals to use.
📝 Technical principles and clinical applications
● Technical principles
1. Detection method: LTA evaluates platelet aggregation by measuring the changes in transmittance of platelet rich plasma (PRP) after the addition of agonists. PRP transmittance is set to 0%, and platelet poor plasma (PPP) is set to 100%. Platelet aggregation can lead to an increase in transmittance.
2. Mechanism of collagen action: Collagen induces platelet aggregation by binding to platelet surface receptors GPIa/IIa and GPVI, initiating adhesion, granule release, and thromboxane A2 production. Due to the dependence on secondary mediators, collagen induced aggregation exhibits a lag phase.
● Clinical application
1. Disease diagnosis: Distinguish between congenital (such as Glanzmann thrombocytopenia) or acquired (such as drug-induced) platelet dysfunction.
2. Drug monitoring: Evaluate the responsiveness of antiplatelet therapy.
🧪Collagen (derived from horse tendon) platelet inducer(Collagen)Reagent composition and preparation
● Reagent components
1. R1: Horse tendon type I collagen (approximately 0.5 mg/bottle), freeze-dried powder, containing stabilizer.
2. R2: Collagen diluent, liquid, non hazardous material.
🎯 Preparation steps
● For gathering instruments:
1. Dissolve R1 in 0.5 mL of distilled water per bottle to 1 mg/mL, shake to dissolve, and stabilize at room temperature (18-25 ° C) for 30 minutes.
2. Prepare 10X working solution: When the final concentration is 5 μ g/mL, take 50 μ L of 1 mg/mL collagen and 950 μ L of diluent.
● Analyzer used:
1. Dissolve each bottle of R1 in 0.625 mL of distilled water to 800 μ g/mL and stabilize at room temperature for 30 minutes.
2. Prepare 8X working solution: When the final concentration is 5 μ g/mL, take 50 μ L of 800 μ g/mL collagen and 950 μ L of diluent.
