1.78PEI MAX 40K transfection reagent(PEI 22K, also known as PEI 22K in free base, is a functional, reliable, and cost-effective transient transfection reagent. In most expression systems of HEK293 and CHO, PEI MAX can provide stable high expression (from 96 well plate to 100L bioreactor). Nowadays, more researchers and companies use PEI MAX for cell transfection every year because compared to most transfection reagents, PEI MAX can achieve stable high expression and reduce transfection costs by 40%.
2.78 PEI MAX 40K is easier to use and has higher transfection efficiency than PEI 25K. Typically, the PEI 25K transfection process takes several hours to prepare, while PEI MAX 40K can complete the entire transfection process within two hours. In addition, PEI 25K contains 4-11% residual acetyl groups, which may hinder the effective binding of the polymer backbone to DNA, while PEI MAX 40K's acetyl groups are prone to detachment, indicating that the performance of each batch of PEI MAX products is more stable.

1. Preparation of storage solution (1 mg/mL)
-Material: 78PEI 40000 powder, ultrapure water, 1N NaOH, 0.1 μ m filter membrane
-Steps:
① Dissolve 1g of powder in 900 mL of ultrapure water and stir magnetically until dissolved;
② Add 1N NaOH dropwise to adjust the pH to 6.9-7.1 (if exceeded, adjust with 1N HCl);
③ Dilute to 1L and filter with a 0.1 μ m membrane for sterilization;
④ After packaging, store at 4 ℃, with a liquid shelf life of 6 months and a freeze-dried powder shelf life of 1 year.
2. Preparation of working fluid
-Dilute according to the ratio of 1 μ g DNA: 1-5 μ L 78PEI 40000 storage solution (recommended initial ratio of 1:3);
-Dilute DNA and PEI with serum-free medium (such as Opti MEM) and let them stand at room temperature for 10-25 minutes to form a complex.
📌 Transfection operation process
(1) Transfection of adherent cells
1. Cell inoculation: Inoculate cells 24 hours before transfection, with a density of 70% -80% confluence, and replace with a culture medium containing 5% serum;
2. Complex incubation: Add DNA-PEI complex dropwise into the culture dish, gently vortex and incubate for 4-6 hours;
3. Fluid exchange and detection: Replace with fresh culture medium, gene expression can be detected after 7 hours, and after 24 hours, subculture and screening drugs can be added.
(2) Suspension cell transfection
1. Cell preparation: Adjust the cell density to 1 × 10 ⁶ cells/mL and culture on a shaker for 2-4 hours;
2. Complex addition: Drop the DNA-PEI complex directly into a shake flask and continuously shake for 24-72 hours;
3. Amplification optimization: After transfection for 24 hours, cells can be diluted (1:2-1:5) to increase protein production.

1. Dissolution requirements:
-It must be fully dissolved, as undissolved particles can lead to a decrease in transfection efficiency;
-Suggest using NEST 5 mL/14 mL centrifuge tubes to handle large volume solutions.
2. pH control:
-The pH should be precisely adjusted to 6.9-7.1 during initial dissolution, as either too high or too low can affect the formation of the complex.
3. Cell state:
-It is recommended to use logarithmic growth phase cells to avoid excessive digestion and damage to the cell membrane.
4. Toxicity control:
-High density cells (>90% confluence) require a reduction in PEI dosage (recommended ratio 1:1).
