During the drug development process, many different formulations of anionic drug encapsulating solutions were screened. Research has found that sucrose octasulfate is a good anionic drug embedding solution. Sucrose octasulfate is added to the finished product production process and reacts to produce triethylammonium sucrose octasulfate (TEA-SOS). Compared with other formulations, this formulation maintains encapsulation efficiency and has a higher retention rate of irinotecan in liposomes.

Potassium sucrose octasulfate(K-SOS) [CDE registration number: F20210000349; AVT item number: 011001, pharmaceutical injection grade, GMP production, bulk stock supply] is a new type of excipient used as raw material to prepare triethylammonium sucrose octasulfate (TEA-SOS) for liposome drug delivery. Sucrose octasulfate is converted to TEA-SOS through ion exchange. TEA-SOS and lipids are passively encapsulated by polycarbonate membrane compression to form liposomes. Subsequently, TEA-SOS outside the liposomes was removed by filtration, forming an electrochemical gradient across the lipid membrane of the liposomes.
