(Experimental Services) Cell Proliferation and Toxicity DeterminationThe core principle is that the CCK-8 reagent contains a compound called WST-8, which is reduced to a water-soluble orange formamide product under the action of dehydrogenase in live cell mitochondria. The higher the cell activity, the stronger the dehydrogenase activity, the more formaldehyde is produced, and the darker the color. The basic operation steps are as follows: Inoculate the cells onto a culture plate and give them different treatments, then directly add CCK-8 reagent and incubate for 1-4 hours. Subsequently, the absorbance values of each well were measured using an enzyme-linked immunosorbent assay (ELISA) reader at a wavelength of 450nm. The OD value shows a good linear positive correlation with the number of live cells, making it possible to accurately quantify cell activity, evaluate compound toxicity, or screen for pro proliferative drugs. This method is safer and more convenient compared to the MTT method, and is widely used in biomedical research.
