How does laser confocal microscope work?

  laser confocal microscopeIt is a high-performance optical microscopy imaging device that combines laser light source and confocal technology. By eliminating non focal plane stray light interference, it achieves subcellular level resolution three-dimensional imaging and is widely used in the fields of life sciences, materials science, and medicine.
  laser confocal microscopeThe core of it is the "confocal optical path" and "point scanning imaging", and the specific process is divided into four steps:
Laser focusing: After being filtered by an excitation filter, the laser light source is focused onto a single tiny point of the sample through an objective lens, which excites the sample to produce fluorescence.
Signal collection: The light emitted by the sample returns along the original optical path, is reflected by a beam splitter, and enters the pinhole aperture (confocal aperture). Only light from the focal point can accurately pass through the pinhole, and stray light from non focal layers (such as the upper and lower layers of the sample) will be blocked by the pinhole.
Point scanning and imaging: The scanning system controls the laser beam to scan the sample surface point by point and line by line, and the optical signal of each point is received by the photodetector and converted into an electrical signal.
3D reconstruction: The computer converts the signal of each scanning point into pixels to form a 2D image; By adjusting the distance between the objective lens and the sample, two-dimensional tomographic images of different depths are obtained, and then the three-dimensional images of the sample are reconstructed by software overlay.
Daily maintenance items: Regularly clean the objective lens (gently wipe with lens paper dipped in lens cleaning solution), avoid direct laser exposure to the eyes (wear specialized laser protective goggles during operation), and keep the sample stage clean (to prevent sample contamination).
