Sample preparation:
Biological samples: Fluorescent labeling (such as labeling target structures with fluorescent dyes or fluorescent proteins) is required to ensure that the samples are dry (or immersed in an appropriate buffer), free of bubbles, and avoid blocking the laser or generating stray light.
Material sample: The surface needs to be cleaned (to remove dust and oil stains). If the sample does not reflect light or has no fluorescence, it can be enhanced by coating (such as spraying gold) to enhance the reflection signal, or dyed with specific dyes.
Instrument operation:
Power on preheating: Turn on the laser light source and computer to preheat 30 Minutes (to ensure stable laser intensity), calibrate the focal point and pinhole position of the objective lens (to ensure confocal effect).
Parameter settings: Select laser wavelength, objective magnification, and scanning speed based on the sample; Adjust the laser intensity and detector gain (to avoid signal saturation or weak blurring); SettingsZAxis scanning range (determining the depth interval of the tomographic scan).
Imaging and saving: Start scanning to obtain 2D or 3D images, perform image processing (such as denoising and pseudocolor coloring) through software, and save the original data and processed images.
Safety precautions: Different wavelengths of lasers have different safety levels; It is prohibited to disassemble the laser module or adjust the optical path without protection to prevent equipment damage or personnel injury.
