In the pharmaceutical industry, water for injection (WFI) is a key raw material for producing sterile drugs such as injections and eye drops, and its quality directly affects the safety and effectiveness of the drugs. Therefore, GMP (Good Manufacturing Practice) has put forward extremely strict requirements for injection water equipment to ensure that the quality of injection water meets the high standards of drug production.
1、 Equipment Material and Design
1. Material selection
The materials used for the storage tanks and pipelines of GMP injection water equipment must be non-toxic and corrosion-resistant, usually made of 316L stainless steel. This material has good corrosion resistance and biocompatibility, which can effectively prevent water pollution. In addition, the inner wall of the equipment needs to be electro polished and passivated to reduce the adhesion of microorganisms and the formation of biofilms.
2. System Design
The pipeline design of the injection water system should avoid dead corners and blind pipes, ensure smooth water flow, and prevent microbial growth. According to GMP regulations, pipelines should adopt a circulation design, and injection water can use insulation circulation above 70 ℃. This design can maintain the turbulent state of water flow (flow velocity ≥ 1.5 m/s), further reducing the risk of microbial contamination.
2、 Water quality monitoring and control
1. Key indicator monitoring
The key quality indicators of injection water include conductivity, total organic carbon (TOC), microbial limits, and endotoxins. GMP requires regular testing and real-time monitoring of these indicators. For example, the conductivity should be ≤ 1.3 μ S/cm (25 ℃), TOC should be ≤ 500 ppb, endotoxin should be ≤ 0.25 EU/mL, and microbial limit should be ≤ 10 CFU/100mL. Through an online monitoring system, these key parameters can be detected in real time to ensure that the water quality meets the requirements.
2. Disinfection and sterilization
In order to prevent microbial contamination, injection water equipment needs to be equipped with effective disinfection and sterilization facilities. Common disinfection methods include high-temperature cycling (70-80 ℃), pure steam sterilization, and ozone disinfection. These methods can effectively kill microorganisms, while verifying the residual risk after disinfection to ensure that it does not cause secondary pollution to water quality.
3、 Verification and maintenance
1. System verification
GMP emphasizes the life cycle validation of injection water equipment, including Installation Qualification (IQ), Operational Qualification (OQ), and Performance Qualification (PQ). These verification steps ensure that the equipment meets the expected quality standards at every stage from installation to operation. In addition, the equipment needs to be revalidated after major changes to ensure its continued effectiveness.
2. Regular maintenance
Regular maintenance is the key to ensuring the stable operation of injection water equipment. The maintenance plan should include regular cleaning, disinfection, replacement of filter membranes/resins, inspection of sealing, etc. Through these measures, the service life of the equipment can be effectively extended while ensuring the stability of water quality.
4、 Document and Record Management
GMP requires pharmaceutical companies to establish a comprehensive document and record management system, covering standard operating procedures (SOPs) for operations, cleaning, disinfection, and other processes. All maintenance activities and water quality monitoring results should be recorded in detail for traceability and auditing purposes. These records not only contribute to the internal quality management of the enterprise, but also serve as an important basis for regulatory inspections.
GMP injection water equipment can effectively meet the strict standards of drug production through strict material selection, system design, water quality monitoring, verification and maintenance, and document record management. Pharmaceutical companies should fully recognize the importance of these requirements and strictly comply with them in practical operations to ensure the quality of injection water and the safety of drugs. Through continuous technological innovation and management optimization, enterprises can improve production efficiency and enhance market competitiveness while ensuring product quality.
